FBXO22、BACH-1、PTEN在复发性流产患者绒毛及蜕膜中的表达研究

摘　　要：目的探讨F-box蛋白22(FBXO22)、BTB-CNC同源体1(BACH-1)及磷酸酯酶及张力蛋白同源基因(PTEN)在复发性流产(RSA)患者绒毛及蜕膜组织中的表达水平。方法选取2022年10月至2023年6月我院产科收治的30例RSA患者作为RSA组,另外选取30例行人工流产的正常早孕患者作为对照组。分别收集两组患者人工流产术后的绒毛和蜕膜组织,采用免疫组织化学法分别检测绒毛及蜕膜组织FBXO22、BACH-1、PTEN的表达定位及水平,采用Western Blot方法检测FBXO22、BACH-1、PTEN在两组患者绒毛组织中的蛋白表达水平。结果免疫组织化学结果显示,FBXO22主要在滋养层细胞和蜕膜基质细胞胞浆中表达,呈棕黄色;BACH-1主要集中在滋养层细胞和蜕膜基质细胞胞核中表达,呈棕黑色颗粒;PTEN主要在滋养层细胞和蜕膜基质细胞胞浆中表达,呈棕黄色。与对照组相比,RSA组绒毛组织和蜕膜组织中FBXO22、BACH-1表达阳性率显著降低(P<0.01),PTEN表达阳性率有升高趋势,但差异无统计学意义(P>0.05)。Western Blot检测结果显示,RSA组BACH-1、FBXO22蛋白的相对表达量显著低于对照组(P<0.01),PTEN蛋白的相对表达量显著高于对照组(P<0.05)。结论FBXO22、BACH-1的表达降低及PTEN的表达升高可能与RSA相关,但仍需要进一步的研究证实。Objective:To explore the expression of F-box protein 22(BXO22),BTB and CNC homology 1(BACH-1),phosphatase and tensin homolog(PTEN)in villi and decidua of patients undergoing recurrent spontaneous abortion(RSA).Methods:Thirty RSA patients admitted to Obstetrics Department of Affiliated Hospital of Shandong Second Medical University from October 2022 to June 2023 were recruited as the RSA group,and another 30 normal early pregnancy patients who underwent induced abortion were selected as the control group.The villi and decidua tissues of two groups were collected after induced abortion,and immunohistochemistry was used to detect the expression localization and levels of FBXO22,BACH-1 and PTEN in the villi and decidua tissues.Western Blot technique was applied to detect the protein expression levels of FBXO22,BACH-1 and PTEN in the villi tissues of the two groups.Results:Immunohistochemical results showed that FBXO22 was mainly expressed in the cytoplasm of trophoblast cells and decidual stromal cells,appearing brownish yellow in color;BACH-1 is mainly expressed in the nuclei of trophoblast cells and decidual stromal cells,appearing as brownish black particles;PTEN is mainly expressed in the cytoplasm of trophoblast cells and decidual stromal cells,appearing brownish yellow in color.Compared with the control group,the positive rates of FBXO22 and BACH-1 expression in the villi and decidual tissues of the RSA group were significantly reduced(P<0.01),and the positive rate of PTEN expression showed an increasing trend,but the difference was not statistically significant(P>0.05).The Western Blot test results showed that the relative expression levels of BACH-1 and FBXO22 proteins in the RSA group were significantly lower than those in the control group(P<0.01),while the relative expression level of PTEN protein was significantly higher than that in the control group(P<0.05).Conclusions:The decreased expression of FBXO22 and BACH-1,as well as the increased expression of PTEN,may be associated with RSA,but further researc
