白刺花总碱对大鼠跟腱损伤的修复作用

摘　　要：为探究白刺花提取的生物总碱(The total alkaloid of Sophora Viciifolia,SVA)对损伤肌腱的修复机制,采用SVA处理大鼠的损伤跟腱,利用跟腱功能指数(Achilles Functional Index,AFI)和组织学评分分析SVA对大鼠跟腱损伤修复的影响。结果表明:SVA显著增加了大鼠的AFI;组织学评分显示SVA处理组肉芽组织、粘连和瘢痕的形成减少;苏木精-伊红(Hematoxylin-eosin staining,H&E)染色结果显示SVA处理组的炎性细胞浸润减少;Masson染色证明SVA处理组比对照组含有更加丰富的胶原纤维;免疫荧光和分子对接等实验证实SVA可以抑制损伤跟腱炎症和血管新生标志因子的高表达,而且在损伤后第14 d加入SVA作用的效果优于在损伤后第7 d加入SVA的效果。研究证明了SVA可以通过调控炎症和血管新生信号通路来促进损伤跟腱的修复。To explore the repair mechanism of the total alkaloid of Sophora Viciifolia(SVA)on injured Achilles tendon,SVA was used to treat the injured Achilles tendon of rats.The effect of SVA on the repair of Achilles tendon injury was analysed by Achilles functional index(AFI)and histological evaluation score.The results showed that SVA treatment significantly increased AFI in rats,and histological observation score showed that the formation of granulation tissue,adhesion and scar were reduced in the SVA treatment group.Hematoxylin-eosin(H&E)staining showed that inflammatory cell infiltration was reduced in the SVA treatment group,and Masson staining proved that the SVA treatment group contained more collagen fibers than the control group,as well as immunofluorescence and molecular docking experiments confirmed that SVA could inhibit the high expression of inflammation and angiogenesis markers of injured Achilles tendon,and the effect of adding SVA on the 14th day after injury was better than that on the 7th day.It is demonstrated that SVA promotes the repair of injured tendons by regulating inflammation and angiogenesis signaling pathways.
