摘  要:目的:本研究旨在探讨急性肺损伤(ALI)后肺纤维化的发病机制,特别是胰岛素样生长因子-1(IGF-1)、转化生长因子β(TGF-β)和血小板源性生长因子(PDGF)在小鼠肺纤维化过程中的作用及其相互关系。方法:使用博来霉素诱导C57BL/6小鼠肺纤维化。通过HE染色和Masson染色评估肺泡炎症和肺纤维化程度,采用Real-time PCR和Western blot技术分析IGF-1、TGF-β和PDGF在mRNA和蛋白水平的表达,并进行相关性分析。结果:实验组小鼠在1~7 d主要表现为肺泡炎症,7~28 d逐渐发展为肺纤维化。IGF-1、TGF-β和PDGF在肺纤维化过程中表达均升高,且三者之间存在正相关关系。IGF-1从始至终参与了肺纤维化过程,而TGF-β和PDGF在早期表达升高,后期有所下降。结论:IGF-1、TGF-β和PDGF在ALI后肺纤维化的发生发展中可能相辅相成,共同促进肺纤维化。Objective To explore the pathogenesis of pulmonary fibrosis following acute lung injury(ALI),particularly the roles and interrelationships of insulin-like growth factor-1(IGF-1),transforming growth factor-β(TGF-β),and plate⁃let-derived growth factor(PDGF)in the pulmonary fibrosis of mice.Methods An ALI-induced pulmonary fibrosis model was established using bleomycin to induce fibrosis in C57BL/6 mice.The alveolar inflammation and pulmonary fibrosis were assessed by hematoxylin-eosin(HE)staining and Masson staining.The mRNA and protein expressions of IGF-1,TGF-β,and PDGF were analyzed using real-time PCR and Western blot,and correlation analysis was performed.Results Mice in the experimental group primarily exhibited alveolar inflammation from days 1 to 7,which progressively developed into pulmonary fibrosis from days 7 to 28.IGF-1,TGF-β,and PDGF were elevated during the process of pulmonary fibrosis,and positive correlations were observed among the three factors.IGF-1 was involved throughout the pulmonary fibrosis,while TGF-βand PDGF increased initially and then decreased.Conclusion IGF-1,TGF-β,and PDGF synergistically promote the occurrence and development of pulmonary fibrosis following ALI.
