摘  要:玉米易受到低温冷害等非生物胁迫侵袭。春季低温冷害会导致东北地区玉米产量降低,挖掘玉米耐冷基因、研究基因功能和分析其调控网络就显得尤为重要。从耐冷玉米自交系W9816中成功克隆到ZmMSRB5(GRMZM2G577677)基因,并将其在拟南芥中异源表达。4℃处理后,ZmMSRB5过表达拟南芥株系(ZmMSRB5-OE)萌发率高于野生型;-8℃冻处理5 h,恢复3 d后,ZmMSRB5-OE株系存活率也明显高于野生型。此外,在4℃条件下,ZmMSRB5-OE株系的PRO含量、SOD和CAT活力都明显高于野生型,丙二醛含量显著低于野生型。说明ZmMSRB5基因可增强拟南芥对冷胁迫的耐受力,在拟南芥响应冷胁迫中起着重要作用。Maize is particularly susceptible to abiotic stresses such as low temperature,which could cause maize production decreased in Northeast China during spring.Therefore,it is particularly important to mine coldtolerant gene,verify the gene function and analyze its regulatory network.In this study,ZmMSRB5(GRMZM2G577677)was successfully cloned from the cold-tolerant maize inbred line W9816 and heterogeneously expressed in Arabidopsis.After 25 days of treatment at 4℃,the germination rate of ZmMSRB5 overexpressing Arabidopsis lines(ZmMSRB5-OE)was higher than WT.After 5 hours of freezing treatment at-8℃and 3 days of recovery,the survival rate of ZmMSRB5-OE lines was also significantly higher than WT.Additionally,the proline content,and the activities of SOD and CAT in ZmMSRB5-OE lines were significantly higher than WT at 4℃,while the MDA content was significantly lower than WT.These findings indicated that ZmMSRB5 could enhance the cold tolerance and play an important role in the response to cold stress in Arabidopsis.
