摘 要:研究表明,MTM1基因的缺失会导致线粒体的功能受到严重损伤,在作物研究中只有零星报告,因而有必要对该基因的生物信息学进行研究。利用前期耐寒基因研究中克隆出花生MEM1基因,得到1275 bp的基因片段,通过生物信息方法对其编码的蛋白质进行分析,该蛋白由424个氨基酸组成,分子量为45843.80 Da,属于不稳定蛋白,无跨膜区域,不属于分泌蛋白;亚细胞定位预测位于细胞膜上;由45.52%的α-螺旋,41.51%的无规则卷曲,4.72%的β-折叠构成二级结构;在花生萌发期胁迫处理下,该基因在耐寒材料中8 h表达量达到最大,在不耐寒材料中24 h表达量达到最大。分析结果为进一步研究该基因的基因功能,以及在花生萌发期耐寒的作用奠定了基础。Recent studies have shown that the loss of the MTM1 gene leads to severe damage to mitochondrial function.However,this gene has only been sporadically reported in crop research,highlighting the need for bioinformatics analysis of this gene.In this study,1275 bp gene fragment encoding a protein was obtained from peanut MEM1 gene cloned in previous cold-tolerance gene research.Bioinformatics methods were used to analyze the protein,which consists of 424 amino acids with a molecular weight of 45843.80 Da.It is an unstable protein without transmembrane regions and does not belong to secretory proteins.Subcellular localization prediction placed it on the cell membrane.The secondary structure is composed of 45.52%α-helix,41.51%random coil,and 4.72%β-sheet.Under stress during peanut germination,the gene expression reached its peak at 8h in cold-tolerant materials and 24 h in non-cold-tolerant materials.This study lays the foundation for further research on the gene function and its role in cold tolerance during peanut germination.
