摘 要:目的探讨四黄消肿散联合消肿活血散外敷对模型小鼠病毒性肺炎的影响。方法将60只小鼠随机分正常对照组(灌胃等体积生理盐水)、模型组(灌胃等体积生理盐水)、奥司他韦组(灌胃75 mg/kg)、四黄消肿散组(外敷)、消肿活血散组(外敷)、联合用药组(外敷+灌胃75 mg/kg),各10只。取乙醚麻醉小鼠,经鼻注射50μL含1个单位半数致死量(LD50)的H1N1病毒以复制流行性感冒病毒性肺炎小鼠模型。正常对照组小鼠经鼻注射等体积生理盐水。建模成功2 h后,灌胃奥司他韦或生理盐水(0.2 mL/10 g);同时,小鼠背部除毛,给予相应药物1.5 g均匀涂抹,并用胶布固定,每天1次,连续1周。称定小鼠肺组织质量并计算肺系数;苏木精-伊红(HE)染色,观察肺组织病理形态;酶联免疫吸附(ELISA)法检测肿瘤坏死因子-α(TNF-α)和白细胞介素6(IL-6)的表达水平;逆转录-聚合酶链反应(RT-PCR)法和Western blot法检测过氧化物酶体增殖物激活受体γ(PPARγ)/核因子κB(NF-κB)信号通路及细胞凋亡相关基因与蛋白的表达水平。结果与模型组比较,奥司他韦组及联合用药组小鼠肺部病理程度均缓解;肺系数显著降低(P<0.05);TNF-α和IL-6水平显著降低(P<0.05);NF-κB,B细胞淋巴瘤-2相关X蛋白(Bax)和半胱氨酸天冬氨酸蛋白酶(Caspase)-3 mRNA及蛋白水平均显著降低,PPARγ和B淋巴细胞瘤-2(Bcl-2)mRNA和蛋白水平均显著升高(P<0.05)。结论四黄消肿散联合消肿活血散外敷可进一步改善模型小鼠肺炎症状,其作用机制可能与抑制PPARγ/NF-κB通路有关。Objective To investigate the effect of external application of Sihuang Xiaozhong Powder combined with Xiaozhong Huoxue Powder on the mice with viral-induced pneumonia.Methods A total of 60 mice were randomly divided into the normal control group(gavage with equal volume of physiological saline),the model group(gavage with equal volume of physiological saline),the oseltamivir group(gavage with 75 mg/kg),the Sihuang Xiaozhong Powder group(external application),the Xiaozhong Huoxue Powder group(external application),and the combination therapy group(external application+gavage with 75 mg/kg),with 10 mice in each group.The mice were anesthetized with ether,and were intranasally injected with 50µL H1N1 virus containing one unit of median lethal dose(LD50)to replicate the mouse model of influenza virus(IFV)-induced pneumonia.The mice in the normal control group were intranasally injected with an equal volume of physiological saline.After 2 h of successful modeling,mice were given oseltamivir or physiological saline by gavage(0.2 mL/10 g);at the same time,1.5 g of the corresponding drugs were evenly applied to the back of the mice after removing hair and fixed with adhesive tape once a day for one week.The lung tissue mass of mice was weighed and the lung coefficient was calculated;hematoxylin-eosin(HE)staining was used to observe the pathological morphology of lung tissue;enzyme-linked immunosorbent assay(ELISA)was used to detect the expression levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6);real-time fluorescence quantitative polymerase chain reaction(RT-PCR)and Western blot were used to detect the peroxisome proliferator-activated receptorγ(PPARγ)/nuclear factor-κB(NF-κB)signaling pathway-related and apoptosis-related genes and proteins.Results Compared with those in the model group,the lung pathological changes of mice in the oseltamivir group and the combination therapy group alleviated;the lung index significantly decreased(P<0.05);the expression levels of TNF-αand IL-6 significantly reduce
