摘 要:目的探讨莪术提取物(榄香烯)对大肠癌HT-29细胞增殖、凋亡的影响及其作用机制。方法CCK-8法检测莪术提取物不同浓度作用下HT-29细胞增殖抑制情况。将HT-29细胞分为实验组1(榄香烯10μmol/L)、实验组2(榄香烯20μmol/L)、实验组3(榄香烯40μmol/L)和对照组(不做处理),并予以相应浓度干预,分别使用克隆形成实验、流式细胞术检测细胞克隆形成能力、细胞凋亡率,使用Western blot检测细胞凋亡蛋白[环氧合酶-2(Cox-2)、髓细胞白血病-1(Mcl-1)、Bcl-2相关X蛋白(Bax)]表达。结果与莪术提取物(0μmol/L)相比,莪术提取物(5μmol/L)作用24 h时的细胞增殖抑制率升高(P<0.05),莪术提取物(10μmol/L)作用8 h、16 h时的细胞增殖抑制率升高(P<0.05);莪术提取物作用8 h、16 h、24 h情况下,HT-29细胞增殖抑制率随着莪术提取物作用浓度逐渐升高(P<0.05)。与对照组HT-29细胞相比,实验组1~3的克隆形成数均降低(P<0.05),细胞凋亡率均升高(P<0.05),Cox-2、Mcl-1蛋白表达均降低(P<0.05),Bax蛋白表达均升高(P<0.05),且存在浓度效应。结论莪术提取物可抑制HT-29细胞增殖,促进凋亡,可能是通过调节Cox-2、Mcl-1和Bax蛋白表达实现对HT-29细胞增殖、凋亡调节。Objective To explore the effects and mechanism of Zedoaria extract(elemene)on proliferation and apoptosis of colorectal cancer HT-29 cells.Methods The proliferation inhibition of HT-29 cells under different concentrations of Zedoaria extract was detected by CCK-8.HT-29 cells were divided into experimental 1 group(elemene 10μmol/L),experimental 2 group(elemene 20μmol/L),experimental 3 group(elemene 40μmol/L)and control group(no treatment),and were intervened at corresponding concentrations.The colony forming ability and apoptosis rate were detected by clone formation assay and flow cytometry,respectively.The expressions of apoptosis related proteins[cyclooxygenase-2(Cox-2),myeloid cell leukemia-1(Mcl-1),Bcl-2 associated X protein(Bax)]were detected by Western blot.Results Compared with Zedoaria extract(0μmol/L),inhibition rate of cells proliferation was increased under Zedoaria extract(5μmol/L)for 24 h(P<0.05),and inhibition rate of cells proliferation was increased under Zedoaria extract(10μmol/L)for 8 h and 16 h(P<0.05).With the increased of Zedoaria extract concentration for 8 h,16 h and 24 h,proliferation inhibition rate of HT-29 cells was gradually increased(P<0.05).Compared with control group,number of clone formation was decreased in all experimental groups(P<0.05),apoptosis rate was increased(P<0.05),expressions of Cox-2 and Mcl-1 proteins were decreased(P<0.05),and expression of Bax protein was increased(P<0.05),showing concentration effect.Conclusion Zedoaria extract can inhibit the proliferation and promote apoptosis of HT-29 cells,which may be related to regulating expressions of Cox-2,Mcl-1 and Bax proteins.
