摘 要:目的通过超高效液相色谱(UPLC)法测定贵州党参的根茎和叶子中党参炔苷的含量。方法采用Waters CORTECS T3色谱柱;流动相为乙腈-水(含0.1%磷酸,比例为23∶77),流速0.4 ml/min;检测波长为268 nm。结果线性方程为y=3.1963×10^(4)x+3.3047×10^(3)(r=0.9993);线性范围0.01081~1.0808μg;根茎中党参炔苷的平均回收率为98.87%(n=6),叶子中党参炔苷的平均回收率为99.39%(n=6)。结论所建立的UPLC法能快速、准确地测定贵州党参的根茎和叶子中党参炔苷含量,重复性好,在实际工作中有较强的实用性。Objective To compare the content of lobetyolin in rhizome and leaf of Codonopsis tubulosa Kom.by ultra-high performance liquid chromatography(UPLC)method.Methods Waters CORTECS T3 chromatographic column was used.The mobile phase was acetonitrilewater(containing 0.1%phosphoric acid,ratio 23:77),flow rate 0.4 ml/min.The detection wavelength was 268 nm.Results The Linear equation was y=3.1963×10^(4)x+3.3047×10^(3)(r=0.9993).The Linear range was 0.01081~1.0808μg.The average recovery rate of lobetyolin in rhizome was 98.87%(n=6).The average recovery rate of lobetyolin in leaf was 99.39%(n=6).Conclusion The established UPLC method can quickly and accurately determine the content of Codonopsis pilosula in the roots and leaves of Codonopsis pilosula from Guizhou,with good repeatability and strong practicability in actual work.
