夹脊穴电针调控TREM2介导的小胶质细胞活化促进脊髓损伤修复的机制研究

摘  要:目的本研究探讨夹脊穴电针通过TREM2调控小胶质细胞的活化对脊髓损伤(spinal cord injury,SCI)大鼠神经元凋亡的机制。方法采用Basso-Beattie-Bresnahan(BBB)和斜坡实验评估大鼠运动功能和平衡功能。TUNEL检测各组大鼠脊髓组织中神经元凋亡。免疫荧光法检测脊髓组织中TREM2的阳性表达。Western blot检测脊髓组织中Iba-1、IL-6、iNOS和Arg-1的蛋白表达。ELISA检测大鼠脊髓组织中炎症因子的含量。结果与SCI组相比,夹脊穴电针治疗后大鼠BBB评分和斜坡角度显著升高,SCI大鼠运动功能和平衡功能有所改善。TREM2的阳性表达及Iba-1、IL-6 and iNOS的蛋白表达降低,Arg-1的蛋白表达增加,神经元凋亡数量减少(P<0.05)。此外,给予TREM2激活剂HSP60干预后,夹脊穴电针对SCI的治疗作用减弱。结论夹脊穴电针治疗可能通过TREM2调控小胶质细胞的活化抑制脊髓组织中神经元细胞的凋亡,改善SCI脊髓损伤,恢复运动功能。Objective To investigate the mechanism of neuronal apoptosis in rats with spinal cord injury through the activation of microglia regulated by TREM2 by electro-acupuncture at jiaji acupoint.Methods The Basso-Beattie-Bresnahan and ramp test were used to assess movement function and balance function in rats.TUNEL assay for neuronal apoptosis in spinal cord tissue of rats in each group.Positive expression of TREM2 in microglia of spinal cord tissue detected by immunofluorescence.Western blot detection of protein expression of Iba-1,IL-6,iNOS and Arg-1 in spinal cord tissue.ELISA for the determination of inflammatory factors TNF-αand IL-1βin rat spinal cord tissue.Results Compared with the SCI group,the BBB score and slope angle were significantly higher in rats treated with chiropractic electroacupuncture,and movement function and balance function were improved in SCI rats.At the same time,positive expression of TREM2 and protein expression of Iba-1,IL-6 and iNOS were reduced,Arg-1 was increased,the number of neuronal apoptosis was significantly reduced(P<0.05).In addition,administration of TREM2 activator intervention resulted in a diminished therapeutic effect of entrapment electrodes on SCI.Conclusion The electroacupuncture at Jiaji Acupoints treatment may inhibit the apoptosis of neuronal cells in the spinal cord tissue through the activation of microglia regulated by TREM2,improving SCI spinal cord injury and restoring movement function.
