摘  要:目的:探讨周末勇士运动(weekend warrior exercise,WWE)对β-肾上腺素能超载诱导心脏损伤的影响及可能机制。方法:将小鼠随机分为对照组(Con组)、模型组[异丙肾上腺素(isoproterenol,ISO)组]和周末勇士运动组(WWE+ISO组)。周末训练持续4周后,采用颈部皮下注射ISO方法建立小鼠心脏损伤模型,3 d后,检测小鼠心脏质量指数(HMI)、血清中肌酸激酶同工酶(CK-MB)、心肌肌钙蛋白I(cTnI)浓度,HE染色和天狼星红染色观察心脏损伤和纤维化程度,TUNEL染色观察心肌细胞凋亡情况,定量PCR、Western blot和免疫荧光染色检测小鼠心脏纤维化和肥大相关基因的表达、炎症相关细胞因子水平以及铁死亡相关基因和蛋白的表达。结果:与ISO组相比,WWE+ISO组HMI值、CK-MB浓度显著降低(P<0.05),cTnI浓度呈现下降趋势(P>0.05)。与ISO组相比,WWE+ISO组心肌细胞排列较为整齐,炎性细胞浸润显著减少,心脏纤维化面积显著减少(P<0.01)。与ISO组相比,WWE+ISO组I型胶原(Col 1)、Ⅲ型胶原(Col 3)和心房利钠肽(Anp)的mRNA水平显著降低(P<0.05),TUNEL阳性细胞数量显著减少(P<0.05)。与ISO组比较,WWE+ISO组白介素6(IL-6)、肿瘤坏死因子α(Tnf-α)和IL-1β的mRNA水平显著降低(P<0.05)。WWE+ISO组核因子E2相关因子2(Nrf2)的mRNA水平、谷胱甘肽过氧化物酶4(GPX4)蛋白表达均显著高于ISO组(P<0.05),WWE+ISO组血红素氧合酶1(HO-1)的mRNA和蛋白表达均明显低于ISO组(P<0.05),WWE+ISO组HO-1荧光阳性细胞数量显著低于ISO组(P<0.05)。结论:周末勇士运动能够减轻β-肾上腺素能超载诱导的心脏损伤,其机制可能与抑制心脏炎症和铁死亡有关。Objective:To explore the effect and possible mechanism of weekend warrior exercise(WWE)onβ‑adrenergic overload induced cardiac injury.Methods:The mice were randomly divided into Con group,ISO group and WWE+ISO group.After 4‑weeks weekend training,the mouse cardiac injury model was established by subcutaneous injection of ISO into the neck.After 3 days,HMI,serum CK‑MB and cTnI concentration were measured.HE staining and Sirius red staining were used to observe the degree of cardiac injury and fibrosis.TUNEL staining was used to observe myocardial cell apoptosis.The qRT‑PCR,Western blot and immunofluorescence staining were used to detect the expression of genes related to cardiac fibrosis and hypertrophy,the levels of inflammation related cytokines and the expression of ferroptosis related genes and proteins in mice.Results:Compared with ISO group,HMI value and CK‑MB concentration in WWE+ISO group significantly decreased(P<0.05),and cTnI concentration showed a downward trend(P>0.05).Compared with ISO group,WWE+ISO group had a more orderly arrangement of myocardial cells and a significant decrease in inflammatory cell infiltration.The area of cardiac fibrosis in WWE+ISO group was significantly lower than that in ISO group(P<0.01).Compared with ISO group,the mRNA levels of Col 1,Col 3 and Anp in WWE+ISO group were significantly reduced(P<0.05).The number of TUNEL positive cells in WWE+ISO group was significantly lower than that in ISO group(P<0.05).Compared with ISO group,the mRNA levels of IL‑6,Tnf‑αand IL‑1βwere significantly reduced in WWE+ISO group(P<0.05).The mRNA levels of Nrf2 and GPX4 protein expression in WWE+ISO group were significantly higher than those in ISO group(P<0.05),the mRNA and protein expression of HO‑1 in WWE+ISO group were significantly lower than those in ISO group(P<0.05).The number of HO‑1 fluorescent positive cells in WWE+ISO group was significantly lower than that in ISO group(P<0.05).Conclusion:Weekend warrior exercise can reduceβ‑adrenergic overload induced card
