摘  要:目的:探讨心理压力应激对小鼠气味选择偏好的影响及相关环路机制。方法:将C57BL/6雄性小鼠随机分为对照组与应激组,应激组结合禁食、束缚应激、昼夜节律颠倒等温和应激刺激及旷场测试建立不可预测温和刺激(UMS)模型,再检测对照组与应激组雄性小鼠选择食物浸出液气味与异性尿液气味的偏好性,利用神经元活动相关的c-Fos表达筛选UMS小鼠在双气味选择实验期间特异激活的候选脑区,并进一步采用化学遗传学操控验证影响UMS小鼠气味选择偏好的关键脑区。结果:对照小鼠对两种气味没有明显的偏好行为,而UMS小鼠对异性尿液表现出显著的偏好行为。在UMS小鼠进行双气味选择偏好实验期间,内侧视前区(MPOA)与中间杏仁核后背侧区(MePD)中的c-Fos阳性神经元数目显著增加。利用化学遗传学激活MePD,雄性小鼠对异性尿液气味表现出偏好;当MePD被抑制时,UMS小鼠对异性尿液气味的偏好显著降低。而MPOA的化学遗传学操控并不影响双选择气味偏好实验的结果。结论:UMS雄性小鼠显著偏好异性尿液的性别信息素气味,而MePD是调控这一行为的关键脑区。Objective:To investigate the effects of psychological stress on odour preference choices of mice and the related neural mechanisms.Methods:C57BL/6 male mice were randomly divided into control group and stress group.An unpredictable mild stress(UMS)model was established in the stress group using mild stress stimuli like fasting,restraint,and circadian rhythm disruption combined with open field tests.Then,the preferences for food extract odour and female urine odour was examined in control and UMS male mice.The specific brain regions activated during dual odour preference tests in UMS mice were identified by the expression of the neuronal activity-related c-Fos.Further,chemogenetic manipulation was used to validate key brain areas influencing UMS mice’s odour preferences.Results:Control mice showed no clear preference for either odour,while UMS mice demonstrated a significant preference for female urine.During the dual odour preference test,an increase in c-Fos positive neurons is observed in the medial preoptic area(MPOA)and posterior dorsal part of medial amygdala(MePD)of UMS mice.Activating MePD resulted in male mice preferring female urine odour,while inhibiting MePD reduced this preference.The manipulation of MPOA did not affect the outcome of the dual-choice odour preference test.Conclusion:UMS male mice has a significant preference for female urine odour,and MePD is a key brain region regulating this behavior.
