摘 要:目的:优选六方藤中总皂苷的提取工艺。方法:利用响应面法对六方藤总皂苷的提取工艺进行优化,以总皂苷提取质量为指标,采用单因素实验法与Box-Benhken响应面法相结合,对提取温度、乙醇浓度、提取时间三因素进行考察,并测定六方藤总皂苷的体外抗氧化活性。结果:六方藤总皂苷最佳提取工艺为提取温度70℃,乙醇浓度74%,提取时间150 min,此条件下提取的六方藤皂苷质量为24.989 mg/g。不同产地的六方藤总皂苷含量存在显著差异,广西河池市宜州区庆远镇的六方藤中的总皂苷含量最高,为29.124 mg/g。结论:优化后的六方藤总皂苷提取工艺稳定、操作简单、重复性良好、可行、所得总皂苷具有较好的抗氧化作用,为其临床开发和应用奠定了基础。Objective:To optimize the best technique for extracting the total Saponins from Cissus hexangularis.Methods:the extraction process of total Saponins from Cissus hexangularis was optimized using response surface methodology.With the extraction yield of total Saponins as the target,the single factor experiment was investigated involving extraction temperature ethanol concentration,and extraction time as factor.A three-factor response surface methodology was developed using the Box-Behnken design,and the antioxidant activities were subsequently determined.Results:The optimal extraction conditions were as follow:extraction temperature 70℃,ethanol concentration 74%,and extraction time 150 min.The extraction yield of total Saponins from Cissus hexangularis was 24.989 mg/g on the optimal condition.The content of total Saponins from different habitats are different,Qingyuan Town is the highest,which is 29.124 mg/g.Conclusion:Cissus hexangularis has high antioxidant activity.When the concentration is 0.08 mg/mL,the scavenging rate of DPPH free radical reaches 80%.The use of response surface method to optimize the extraction process of total Saponins from Cissus hexangularis s stable,simple,reproducible and feasible.had the accuracy and reliability,and the total Saponins obtained had a strong antioxidant activity,which lays a foundation for its clinical development and application.
