摘  要:目的:提取高浓度再生丝素蛋白溶液,为再生丝素蛋白溶液的制备提供有价值参考。方法:将天然蚕丝经碳酸钠溶液高温脱胶、氯化钙-乙醇-水三元体系(摩尔比1∶2∶8)溶解,之后依次经过透析、浓缩、离心、过滤等多个步骤处理,以获取再生丝素蛋白溶液。结果:通过上述方法提取出的再生丝素蛋白溶液呈现出明亮的金黄色,浓度约为14.9wt%;同时,运用傅里叶变换红外光谱技术对提取的丝素蛋白进行表征,结果显示其具备典型的丝素蛋白结构特征。结论:本研究成功开发出一种基于氯化钙-乙醇-水三元体系的再生丝素蛋白溶液制备方法,该方法能够满足多个领域的需求。Objective:To extract high concentration regenerated silk fibroin protein solution and provide a valuable reference for the preparation of regenerated silk fibroin protein solution.Methods:Natural silk was degelatinized in sodium carbonate solution at high temperature,dissolved in calcium chloride-ethanol-water terpolymer system(molar ratio 1∶2∶8),and then treated by dialysis,concentration,centrifugation and filtration in order to obtain the regenerated silk fibroin protein solution.Results:The solution of regenerated silk fibroin protein extracted by the above method showed bright golden color,the concentration was about 14.9 wt%.At the same time,the extracted silk fibroin protein was characterized by Fourier transform infrared spectroscopy,and the results showed that it had typical structural characteristics of silk fibroin protein.Conclusion:In this study,a method for preparing regenerated silk fibroin protein solution based on calcium chloride-ethanol-water terpolymer system has been successfully developed,which can meet the needs of many fields.
