当归与川芎茎叶精油缓解UV诱导细胞光损伤的作用研究

摘  要:为了确定当归与川芎茎叶精油的利用价值。本研究通过CCK8法测定两种精油的安全浓度范围,采用紫外线A(UVA)、紫外线B(UVB)照射,诱导人永生化角质形成细胞(HaCaT)和人皮肤成纤维细胞(HSF)形成光损伤,并以细胞活力和细胞内活性氧(reactive oxygen species,ROS)的含量作为评价指标,确定细胞的损伤程度;以细胞活力和细胞超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPX)及乳酸脱氢酶(LDH)的含量来评估精油抗细胞光损伤的作用效果。结果表明:两种精油的安全实验浓度最大值均为400μg/mL(HaCaT细胞),川芎叶精油为400μg/mL、当归叶精油为500μg/mL(HSF细胞);细胞光损伤模型中,UVA诱导HaCaT细胞光损伤分别选取12.528 mJ/cm^(2)、15.66 J/cm^(2)、UVA诱导HSF细胞光损伤分别选取3.132 mJ/cm^(2)、18.792 J/cm^(2)、UVB诱导HaCaT细胞光损伤分别选取15 mJ/cm^(2)、60 mJ/cm^(2)、UVB诱导HSF细胞光损伤分别选取150 mJ/cm^(2)、600 mJ/cm^(2)作为低损伤和高损伤的照射剂量;在UV低剂量照射时,精油的修复效果较好,最高可以提升24%左右的细胞活力;对于HaCaT细胞,低浓度1μg/mL川芎叶精油的修复效果普遍高于当归,表现为细胞活力、抗氧化酶含量与细胞膜完整性的提高,对于HSF细胞,在UV低剂量损伤下中浓度250μg/mL当归叶精油效果最佳。在适宜的精油浓度下,相比于模型组,精油处理后细胞活性提升、抗氧化酶含量增加、细胞膜完整性增强,可见两种精油对细胞光损伤具有一定的缓解功效。因此,当归和川芎废弃叶中的精油具有用于抗皮肤光损伤产品开发的潜力。To determine the application value of Angelica sinensis and Ligusticum chuanxiong stem and leaf essential oils.Cell Counting Kit-8(CCK8)assay was used to evaluate essential oil safety upper limit in this study.Human immortalized keratinocyte(HaCaT)and human fibroblast(HSF)were exposed to UVA/B to induce cellular photo-damage,which was weighed using cell viability and levels of reaction oxygen species(ROS).Cell Counting Cell viability and content of superoxide dismutases(SOD),catalase(CAT),glutathione peroxidase(GPX)and lactate dehydrogenase(LDH)were detected to assess the anti-photodamage effect of two essential oils.The safety evaluation experiment suggested an maximum experimental concentration of 400µg/mL for both essential oils on HaCaT,and 500µg/mL and 400µg/mL for A.sinensis and L.sinense‘chuanxiong’,respectively,on HSF.In the cell photodamage model,low-and high-dose UVA,which were 12.528 J/cm^(2) and 15.66 J/cm^(2) for HaCaT,3.132 J/cm^(2) and 18.792 J/cm^(2) for HSF,and low-and high-dose UVB,which were 15 mJ/cm^(2) and 60 mJ/cm^(2) for HaCaT,150 mJ/cm^(2)and 600 mJ/cm^(2) for HSF,were used to induce low and high degree cellular damage.Overall,essential oils revived cells under low-dose UV radiation with a maximum of 24%enhancement in cell viability.Low concentration of L.sinense‘chuanxiong’essential oil(1µg/mL)exhibited better restorative effect than A.sinensis on HaCaT as reflected by elevated cell viability,anti-oxidative enzyme activities and cell membrane integrity.For HSF,under low-dose UV radiation,A.sinensis essential oil at middle concentration(250µg/mL)exhibited the best performance.Overall,treatment with essential oils at appropriate concentrations enhanced cell viability,anti-oxidative enzyme activities,and cell membrane integrity when compared to the model,indicating two essential oils showed certain ameliorating effect on cell photodamage.Therefore,essential oils extracted from the abandoned A.sinensis and L.sinense‘chuanxiong’leaves can be potentially developed into products
