摘 要:[目的]本研究对香榧WOX基因家族进行鉴定并分析了体细胞胚发生相关WOX表达,以筛选香榧体细胞胚发生关键WOX基因,提升香榧体细胞胚发生效率,以期解决香榧育种工作中生长周期长的困难。[方法]用拟南芥WOX蛋白序列比对香榧基因组,鉴定出的WOX基因进行蛋白理化性质、保守基序、保守结构域和染色体定等分析,进一步分析了香榧根、茎、叶、愈伤和体细胞胚中的WOX基因的表达量。[结果]本研究共鉴定出17个香榧WOX成员,系统进化分析后将其分为3个进化支:WUS进化支、中间进化支和古老进化支;不同进化支上的基因均含有特异性的保守结构域;17条基因定位在6条染色体上,其理化性质也各有不同;香榧的WOX2、WOX8、WOX9、WOX13在根、茎、叶、体胚、愈伤上均有不同程度的表达,并且在体细胞胚和愈伤中的表达量较高。[结论]基于香榧WOX2、WOX8B、WOX9A、WOX13A~D在体细胞胚和愈伤中的高表达量,推测它们可能参与香榧愈伤的形成与体细胞胚的发育。[Objective]The WOX(Wuschel-related homeobox)gene family is a unique class of plant-specific transcription factors that serve as a stem cell marker genes and play crucial roles in somatic embryogenesis.Torreya grandis,a conifer species known for producing economically valuable nuts,faces challenges in breeding due to its prolonged growth cycle.This study aims to identify the members of the WOX gene family in Torreya grandis and analyze their expression patterns to identify key WOX genes involved in somatic embryogenesis.The findings are expected to contribute to enhance understanding of the WOX genes’roles,improve the efficiency of somatic embryogenesis,and support the breeding.[Method]Based on the genome data of Torreya grandis,the WOX genes were identified,and heir physicochemical properties,conserved motifs,conserved domains,and chromosome localization were analyzed.Additionally,we examined the expression patterns of these WOX genes across various tissues,including roots,stems,leaves,callus,and somatic embryos.[Result]A total of 17 WOX family members were identified,which were classified into three phylogenetic clades:the WUS clade,the intermediate clade,and the ancient clade.Each clade contained genes with unique conserved domains.These genes were distributed across six chromosomes.Notably,WOX2,WOX8,WOX9 and WOX13 showed variable expression in roots,stems,leaves,callus and somatic embryos,with notably higher expression levels in somatic embryos and callus.[Conclusion]The relatively higher expression of WOX2,WOX8,WOX9 and WOX13 in somatic embryos and calluses suggests that these genes play significant roles in the initiation and maintenance of embryogenic callus and the development of somatic embryos in Torreya grandis.
