摘 要:目的研究四神丸含药血清对结肠癌HT29细胞的抑制作用。方法将HT29细胞分为Control组(加入正常小鼠血清)、四神丸含药血清组(SSW)及FBS组(加入10%胎牛血清)。SSW分别采用2.5%、5%、10%的四神丸含药血清对结肠癌HT29细胞进行干预,通过CCK-8检测四神丸含药血清对HT29细胞增殖的影响;细胞划痕实验检测HT29细胞迁移;Hoechst33342染色检测HT29细胞凋亡;免疫荧光法检测HT29细胞凋亡相关因子Bax的表达;Western blot法检测HT29细胞中NF-κBp65、STAT3、p-NF-κBp65、p-STAT3和TNF-α的表达。结果与Control组相比,10%SSW组的细胞活力显著降低(P<0.01),给药组的划痕距离明显大于Control组(P<0.01);Hoechst33342染色显示四神丸组细胞凋亡增加;SSW组的促凋亡蛋白Bax荧光强度明显高于对照组;给药组的蛋白表达水平明显低于对照组。结论四神丸含药血清可能通过抑制NF-κB、STAT3信号通路的激活而抑制HT29细胞的增殖及迁移能力,诱导HT29细胞凋亡。Objective To study the inhibitory effect of serum containing Sishenwan on HT29 cells.Methods HT29 cells were divided into a blank control group(administered with normal mouse serum),a group treated with Sishenwan(administered with Sishenwan-containing serum),FBS group(administered with 10%fetal bovine serum).The administration group of Sishenwan received intervention with 2.5%,5%and 10%Sishenwan-containing serum on colon cancer HT29 cells,and the effect of Sishenwan-containing serum on HT29 cell proliferation was detected by CCK-8;HT29 cell migration was detected by cell scratch assay.The apoptosis of HT29 cells was detected by Hoechst33342 staining.The expression of apoptosis-related Bax in HT29 cells was detected by immunofluorescence method.The expression of NF-κBp65,STAT3,p-NF-κBp65,p-STAT3 and TNF-αin HT29 cells was detected by Western blot.Results Compared with the control group,the cell viability of 10%Sishenwan-containing serum group was significantly decreased(P<0.01),and the scratch distance of the drug administration group was significantly greater than that of the control group(P<0.01).Hoechst33342 staining showed an increase in cell apoptosis in the Sishenwan group.The fluorescence intensity of pro-apoptotic Bax in the administration group was significantly higher than that in the control group.The protein expression level of the administration group was significantly lower than that of the control group.Conclusion The Sishenwan-containing serum might inhibits the proliferation and migration ability of HT29 cells,promote apoptosis of HT29 cells by inhibiting the activation of NF-κB and STAT3 signaling pathway.
