摘  要:目的研究并筛选亳菊的抗氧化活性成分,初步分析其药效物质基础。方法以亳菊乙酸乙酯提取物和从亳菊中分离得到的16种黄酮类化合物为材料,采用DPPH自由基、ABTS自由基和超氧阴离子(DPPH·、ABTS·^(+)、O^(2-))清除法对抗氧化活性进行评价,并对其黄酮类化合物抗氧化活性与结构之间的关系进行了分析;以芦丁为对照品测定了提取物中总黄酮的含量。结果亳菊乙酸乙酯部位总黄酮含量为14.54%;亳菊中乙酸乙酯提取物以及黄酮类化合物对各种自由基均具有一定的清除能力,在一定浓度范围内抗氧化作用与浓度呈剂量依赖性,随着浓度增加抗氧化活性相应增强。浓度为1.0 mg/mL时,乙酸乙酯提取物抗氧化活性与阳性对照组维生素C非常接近。黄酮类化合物活性可能与B环上羟基的数量和位置有关;而C环上的3位羟基、2位、3位的双键也对活性有一定程度的影响。结论亳菊中黄酮类化合物以及乙酸乙酯提取物对各种自由基均具有一定的清除能力。抗氧化作用与它们的浓度呈明显的量效关系。Objective To study and screen the anti-oxidant active constituents of Chrysanthemum morifolium(Ramat.)Tzvel.‘Boju’,a preliminary investigation on the material basis of its medicinal effects was carried out.Methods The ethyl acetate extract and 16 flavonoids isolated from Boju were examined for their scavenging ability against DPPH radicals,ABTS radicals and superoxide anions(DPPH·,ABTS·^(+),O^(2-)),and the relationship between the anti-oxidant activity of the flavonoids and their structure was analyzed.The content of total flavonoids in the extract was also determined using rutin as a control.Results The content of total flavonoids in the ethyl acetate part of Boju was 14.54%;the ethyl acetate extract and the flavonoids had a certain scavenging ability for various free radicals,and the antioxidant effect was dose-dependent with the concentration in a certain concentration range,and the anti-oxidant activity was enhanced with the increase of the concentration accordingly.At a concentration of 1.0 mg/mL,the antioxidant activity of ethyl acetate extract was very close to that of the positive control Vc.The flavonoid activity may be related to the number and position of hydroxyl group on the B ring;while the 3-position hydroxyl group on the C ring,and the double bonds at the 2-position and 3-position also affected the activity to a certain extent.Conclusion The flavonoids in Chrysanthemum morifolium(Ramat.)Tzvel.‘Boju’as well as the ethyl acetate extract have a certain scavenging ability for various free radicals.The anti-oxidant effects show a significant quantitative relationship with their concentrations.
