摘  要:目的研究引火汤对绝经后抑郁症模型小鼠突触可塑性的影响及作用机制。方法实验分为Sham组、OVX+CUMS组、E_(2)组和引火汤组。通过卵巢切除联合慢性不可预见性轻度应激复制绝经后抑郁症模型,每日灌胃给药1次,连续28 d。悬尾和蔗糖偏好实验评估小鼠抑郁样行为;HE染色观察海马神经元形态及子宫形态;高尔基染色观察树突棘形态及数量;Western blot、RT-qPCR检测海马突触可塑性相关指标;ELISA检测线粒体功能及能量代谢相关指标。结果与OVX+CUMS组比较,E_(2)和引火汤组小鼠不动时间显著降低、糖水偏好指数显著增加,神经元形态和树突棘密度正常,E_(2)组子宫各层结构完整,UC显著性增加;YHT组UC无显著性差异;海马PSD95、NR2A、NR2B、KIF17、ERβ和NRF-1表达增加;LDH活性降低、PDH、α-KGDH、COXⅣ活性升高;E_(2)和ATP水平增高。结论引火汤可改善OVX+CUMS小鼠抑郁样行为,并增强其突触可塑性,效果与E_(2)组相当,其机制可能是通过雌激素效应,改善海马区线粒体功能、调节葡萄糖代谢实现的。Objective To study the effect and mechanism of Yinhuo Tang on synaptic plasticity in postmenopausal depression model mice.Methods The experiment was divided into Sham group,OVX+CUMS group,E_(2)group and Yinhuo Tang group.A postmenopausal depression model was replicated by ovarian resection combined with chronic unpredictable mild stress,administered orally once a day for 28 consecutive days.Evaluation of depression like behavior in mice using tail suspension and sucrose preference experiments;HE staining was used to observe the morphology of hippocampal neurons and uterine morphology;Gorky staining was used to observe the morphology and quantity of dendritic spines;Western blot and RT-qPCR were used to detect hippocampal synaptic plasticity related indicators;ELISA was used to detect mitochondrial function and energy metabolism related indicators.Results Compared with the OVX+CUMS group,the E_(2)and Yinhuo Tang groups showed a significant decrease in immobility time,a significant increase in sugar water preference index,normal neuronal morphology and dendritic spine density,and intact uterine layer structure in the E_(2)group,with a significant increase in UC;There was no significant difference in UC between the Yinhuo Tang group and the control group;The expression of PSD95,NR2A,NR2B,KIF17,ERβand NRF-1 in the hippocampus increased;LDH activity decreased,while PDH,α-KGDH and COX IV activity increased;The levels of E_(2)and ATP have increased.Conclusion Yinhuo Tang can improve depression like behavior in OVX+CUMS mice and enhance their synaptic plasticity,with a similar effect to the E_(2)group.Its mechanism may be achieved through estrogen effects,improving mitochondrial function in the hippocampus and regulating glucose metabolism.
