摘 要:目的鉴定三七HSF(PnHSF)家族,并分析其基因结构、蛋白结构及表达模式。方法采用生物信息学方法,从三七基因组文件中鉴定出PnHSF基因,对其蛋白理化性质、亚细胞定位、保守基序(motif)、启动子顺式元件、蛋白互作网络、染色体定位以及表达模式进行分析。结果共鉴定出24个PnHSF基因,均具有HSF-box结构域。PnHSF蛋白中鉴定出10个保守基序,基因由1个及以上的外显子组成。PnHSF启动子含有多个激素响应元件及MYB结合位点。PnHSF家族成员分布于9条染色体上。表达分析发现PnHSF基因在芦头与主根的表达量大于叶片,摘蕾会引起大部分PnHSF基因在芦头与主根中的表达降低。结论本研究对PnHSF转录因子家族进行全基因组鉴定,共鉴定出24个PnHSF基因。芦头与主根的表达量大于叶片,摘蕾后大部分PnHSF基因表达量下降,推测PnHSF家族可能参与调控三七皂苷的生物合成。Objective To identify the Panax notoginseng HSF(PnHSF)family and analyze its gene structure,protein structure and expression pattern.Methods A bioinformatics approach was used to identify the PnHSF gene from the Panax notoginseng genomic file,and its protein physicochemical properties,subcellular localization,conserved motifs,promoter cis-elements,protein interactions network,chromosomal localization,and expression pattern were analyzed.Results A total of 24 PnHSF genes were identified,all of which had the HSF-box domain.10 conserved motifs were identified in the PnHSF protein,and the gene consisted of 1 or more exons.the PnHSF promoter contained several hormone response elements and MYB transcription factor binding sites.the PnHSF family members were distributed on 9 chromosomes.Expression analysis revealed that the expression of PnHSF genes was greater in root-head and taproot than that in leaves,and inflorescence removal caused a decrease in the expression of most of the PnHSF genes in root-head and taproot.Conclusion In this study,a genome-wide characterization of the PnHSF transcription factor family is performed,and 24 PnHSF genes are identified.The expression of root-head and taproot is greater than that of leaves,and the expression of most PnHSF genes decreased after inflorescence removal.The results suggested that the PnHSF family may be involved in the regulation of Panax notoginseng saponin biosynthesis.
