银杏中总黄酮的超声提取及含量测定

摘  要:研究以银杏叶和银杏果为原料,采用超声法提取银杏总黄酮,通过紫外-分光光度法测定单因素实验条件下银杏中不同部位总黄酮的含量,寻求银杏叶和银杏果中黄酮提取的最佳工艺条件。银杏叶中黄酮超声提取的最佳条件为:乙醇浓度60%,料液比为1:40(g/mL),粉末粒度为80目,提取时间为60 min,提取温度为70℃,在此条件下银杏叶中黄酮提取率为2.44%;银杏果黄酮超声提取的最佳条件为:乙醇浓度60%,料液比为1:40(g/mL),粉末粒度为45目,提取时间为75 min,提取温度为60℃,在此条件下银杏果中黄酮提取率为0.83%。在最佳提取条件下,银杏叶和银杏果中黄酮提取率较为稳定,银杏叶中黄酮含量显著高于银杏果中黄酮含量,为银杏中黄酮的药用价值开发提供依据和参考。The study used Ginkgo biloba leaves and Ginkgo biloba fruits as raw materials to extract the total flavonoids of Ginkgo biloba by ultrasound.The content of total flavonoids in different parts of ginkgo biloba was determined by uv-Spectrophotometer under single-factor experimental conditions,to search for the best technological conditions of extracting flavonoids from Ginkgo biloba leaves and Ginkgo biloba fruit.The optimum conditions of ultrasonic extraction of flavonoids from Ginkgo biloba leaves were as follows:ethanol concentration 60%,solid-liquid ratio 1:40(g/mL),particle size 80 mesh,extraction time 60 min,extraction temperature 70℃,under these conditions,the extraction rate of flavonoids from Ginkgo biloba leaves was 2.44%,and the optimum conditions for ultrasonic extraction of flavonoids from Ginkgo biloba fruits were as follows:ethanol concentration 60%,ratio of solid to liquid 1:40(g/mL),particle size 45 mesh,extraction time 75 min,the extraction temperature was 60℃,and the extraction rate was 0.83%.Under the optimum extraction conditions,the extraction rate of flavonoids in Ginkgo biloba leaves and ginkgo biloba fruit was relatively stable,and the content of flavonoids in Ginkgo biloba leaves was significantly higher than that in Ginkgo biloba fruit.
