摘  要:目的 研究长期暴露石油化工源性PM2.5对雄性大鼠下丘脑-垂体-性腺轴相关激素与形态学结构的影响,探讨石油化工来源性PM2.5对生殖相关器官功能和形态异常发展中的机制。方法 40只8周龄雄性SD大鼠经性功能和勃起功能试验确认有正常性功能后,随机选取10只作为正常组,其余30只进行造模,随机分为造模组15只和造模后组15只,采用智能综合大气采样器将石油化工厂PM2.5过滤至玻璃纤维过滤膜并制成0.65 mg/l的混悬液经鼻染毒,滴注至大鼠气管内,每周染毒3次;正常组大鼠经鼻滴注同体积生理盐水,3组大鼠每周剂量体质量并通过交配试验观察其性功能变化,经每周性功能试验结果对比后,确定造模成功,并第9周处死正常与造模组检测其体质量、睾丸系数指标和外周血清中性激素的水平,镜下观察苏木精-伊红(HE)染色睾丸组织学结构变化。造模后组在正常环境中再饲养2周后处死,检测体质量、睾丸系数指标和外周血清中性激素的水平并镜下观察睪丸组织结构变化。结果 与正常组大鼠相比,造模组与造模后组大鼠体质量增长明显缓慢(P<0.05),造模后组与造模组体质量变化差异无统计学意义(P>0.05)。与正常组比较,造模组与造模后组大鼠性功能明显变弱(P<0.05),造模组与造模后组大鼠性功能差异无统计学意义(P>0.05)。与正常组比较,造模组与造模后组大鼠外周血中的促性腺激素释放激素(GnRH)和雌激素(E)水平降低,但差异无统计学意义(P>0.05),促卵泡激素(FSH)、雄激素(T)和促黄体生成素(LH)明显降低(P<0.05),催乳素(PRL)明显升高(P<0.05),雌激素水平均值降低但无统计学意义(P>0.05),造模组与造模后组以上性激素指标均无统计学意义(P>0.05)。通过睪丸组织重量对比,与正常组大鼠相比,造模组与造模后组大鼠睪丸重量明显减少(P<0.05),造模组与造模后组睾丸重量差异无统计学意义(P>0.05)�Objective To investigate the effects of long⁃term exposure to petrochemical⁃derived PM2.5 on hy⁃pothalamic⁃pituitary⁃gonadal axis⁃related hormones and morphological structure in male rats,and to explore the mech⁃anism of petrochemical⁃derived PM2.5 on the abnormal development of reproductive organs.Methods 408⁃week⁃old male SD rats with normal sexual function confirmed by sexual function and erectile function test were ran⁃domly selected as the normal group(n=10),and the other 30 were selected as the modeling group,and after modeling group(n=15).An intelligent comprehensive air sampler was used to filter PM2.5 from A petrochemical plant into a glass fiber filter membrane and make a suspension of 0.65 mg/l into the trachea of rats for nasal poisoning three times a week.The rats in normal group were injected with the same volume of normal saline through the nose.The rats in the three groups were measured weekly and their sexual function changes were observed through mating experiment.After comparing the results of the sexual function experiment every week,the formation of the mold was confirmed,and the body weight,testicular coefficient index and sex hormone level in peripheral blood of normal group and modeling group were detected at the 9th week,and the morphological structure changes of testis were observed under the micro⁃scope.The after modeling group was fed in a normal environment for another two weeks and killed.The body weight,testicular coefficient and sex hormone levels in peripheral blood were detected.The morphological and structural changes of testicle were observed by HE staining.Results Compared with normal group,the body weight of rats in modeling group and after modeling group increased slowly(P<0.05),and there was no difference in body weight be⁃tween after modeling group and modeling group(P>0.05).Through the mating experiment,compared with normal group,the sexual function of rats in modeling group and after modeling group was significantly weakened(P<0.05),and the sexu
