桑叶中山柰酚的分离鉴定及细胞毒活性研究

摘  要:桑叶乙醇提取物的乙酸乙酯萃取部位经硅胶柱层析和中压制备色谱得到一淡黄色粉末,经^(1)H-NMR和^(13)C-NMR数据分析,确定其化学结构为山柰酚。利用MTT法检测其对小鼠乳腺癌细胞株4T-1、人口腔表皮癌细胞株KB、小鼠肺癌细胞株LLC和人大细胞肺癌细胞株NCl-H1299的细胞毒活性。结果表明,山柰酚作用于4T-1和LLC细胞48 h的IC_(50)值分别是(15.65±0.66)μg/mL和(22.54±1.09)μg/mL;山柰酚作用于KB和NCI-H1299细胞48 h的IC_(50)值分别是(47.26±3.25)μg/mL和(39.20±5.75)μg/mL。山柰酚对4T-1和LLC两种瘤株有较强抑制作用,对KB和NCl-H1299细胞增殖抑制作用较弱。山柰酚为桑叶抗肿瘤活性成分之一。The ethyl acetate extract of Mori folium was subjected to silica gel column chromatography and medium pressure preparation chromatography to obtain a light-yellow powder.After analysis of ^(1)H-NMR and ^(13)C-NMR data,its chemical structure was determined to be kaempferol.The cytotoxicity against 4 cancer cell lines was evaluated by MTT assay.The results showed that the IC_(50) values of kaempferol acting on 4T-1 and LLC cells for 48 h were(15.65±0.66)μg/mL and(22.54±1.09)μg/mL,respectively.The IC_(50) values of kaempferol acting on KB and NCI-H1299 cells for 48 h were(47.26±3.25)μg/mL and(39.20±5.75)μg/mL,respectively.Kaempferol,a flavonoid component from Mori folium has a certain inhibitory effect on the proliferation of mouse breast cancer cell 4T-1 and mouse lung cancer cell LLC,and it is one of the antitumor active ingredients of Mori folium.
