摘  要:目的:研究铁皮石斛多糖对动脉粥样硬化(atherosclerosis,AS)大鼠血脂和相关影响因子的影响,探究其抗动脉粥样硬化的作用机制。方法:给予动脉粥样硬化大鼠高、中、低剂量铁皮石斛多糖,9周后处死大鼠,取血测定TC、TG、LDL-C;主动脉油红O染色,进行病理形态学观察;测定血清中TNF-α、CRP、VEGF和G-CSF因子水平。结果:与AS组相比,DPH组、DPM组、DPL组主动脉粥样斑块面积均有不同程度缩小,脂质积累明显减少,DPH组和DPM组大鼠血清TC、TG及LDL-c水平明显降低。与AS组相比,铁皮石斛多糖DPH组、DPM组、DPL组大鼠血清中TNF-α、C-CRP、VEGF水平均有不同程度降低,G-CSF水平提高,且铁皮石斛多糖药效呈浓度依赖性。结论:铁皮石斛多糖能有效降低动脉粥样硬化大鼠血清中有关脂质含量,降低血清内TNF-α、CRP、VEGF的表达,提高G-CSF水平,减少动脉粥样硬化的发生。Objective To study the effects of Dendrobium officinale polysaccharide on blood lipids and related factors in atherosclerosis(AS)rats,and to explore its mechanism of action against atherosclerosis.Methods Dendrobium dendrobium polysaccharide was given to atherosclerotic rats at high,middle and low doses,and the rats were killed 9 weeks later,and TC,TG and LDL-C were determined by blood collection.Aorta oil red O staining was performed to observe the pathomorphology.Serum levels of TNF-α,CRP,VEGF and G-CSF were determined.ResultsCompared with AS group,aortic atherosclerotic plaque area was reduced to varying degrees in DPH group,DPM group and DPL group,lipid accumulation was significantly reduced,and serum TC,TG and LDL-c levels in rats in DPH group and DPM group were significantly reduced.Compared with AS group,the levels of TNF-α,C-CRP and VEGF in serum of rats in D.officinale polysaccharides DPH,DPM and DPL groups were decreased to varying degrees,and the level of G-CSF was increased,and the pharmacodynamics of D.officinale polysaccharides was concentration-dependent.Conclusion Dendrobium officinale polysaccharides can effectively reduce the content of related lipids in serum of atherosclerotic rats,reduce the expression of TNF-α,CRP and VEGF in serum,increase the level of G-CSF,and reduce the occurrence of atherosclerosis.
