摘 要:为探究高山红景天、枸杞、荷叶及五味子降糖颗粒剂的制备工艺。本研究采用湿法制粒,以颗粒剂的外观形状,粘度等为评价指标进行单因素试验,筛选出微晶纤维素和甘露醇的最佳辅料重比,根据其日用量设计L9(34)正交试验表,按《中国药典》2020年版进行质量检查。使用紫外可见光分光光度法测其总多糖、总黄酮、总生物碱及总木脂素含量。辅料选择微晶纤维素做填充剂,甘露醇做矫味剂,最佳辅料重量比为1:1,制得的颗粒剂均符合《中国药典》2020年版质量检查标准。结合降糖药理试验,优化得到降糖效果最好的主药组合为G8组,最佳降糖日用量为高山红景天6 g,枸杞9 g,荷叶3 g,五味子2 g。本研究优选出的工艺简便可行,为今后降糖颗粒剂的研究提供理论参考依据。To explore the preparation process of Rhodiola cretinii,Lycium barbarum,Nelumbo nucifera and Schisandra chinensis hypoglycemic granules.In this study,single factor experiment was carried out with the appearance,shape and viscosity of the granules as the evaluation indexes to select the best weight ratio of microcrystalline cellulose and mannitol.The L9(34)orthogonal experiment table was designed according to the daily dosage,and the quality inspection was carried out according to the Chinese Pharmacopoeia 2020 edition.The content of total polysaccharides,total flavonoids,total alkaloids and total lignans in the granules were determined by UV.Microcrystalline cellulose was chosen as the filling agent and mannitol as the flavoring agent.The best weight ratio of the excipients was 1:1.The prepared granules all met the quality inspection standards of the 2020 edition of the Chinese Pharmacopoeia.Based on hypoglycemic pharmacological experiments,the G8 group was optimized to have the best hypoglycemic effect.The optimal daily hypoglycemic dosage was 6 g of R.cretinii,9 g of L.barbarum,3 g of N.nucifera,and 2 g of S.chinensis.The optimized process was simple and feasible,providing a theoretical reference basis for future research on hypoglycemic granules.
