摘 要:本研究旨在明确人参皂苷CK通过对Nrf2/Keap1通路基因的调控,改善小鼠认知障碍和神经元保护作用的分子机制研究。本研究通过腹腔注射氢溴酸东莨菪碱(SCOP)创建小鼠认知障碍模型,通过水迷宫系统检测各组小鼠的记忆和学习能力,通过尼氏染色观察脑组织神经元尼氏体数量和形态,采用试剂盒检测血清中GSH含量,利用实时荧光定量PCR检测Nrf2/Keap1信号通路和凋亡相关基因的表达水平。结果表明,人参皂苷CK提高认知障碍小鼠学习记忆能力,抑制小鼠脑组织海马神经元损伤,提高血清内GSH含量,调节脑内Nrf2/Keap1信号通路相关基因的表达,抑制脑组织细胞凋亡。人参皂苷CK可通过调控Nrf2/Keap1信号通路相关基因,抑制小鼠脑组织神经元氧化损伤和凋亡,改善小鼠认知障碍。To clarify the molecular mechanism by which ginsenoside CK ameliorates cognitive impairment and neuronal protection in mice through the regulation of Nrf2/Keap1 pathway genes.In this study,a mouse model of cognitive impairment was established by intraperitoneal injection of scopolamine hydrobromide(SCOP).Memory and learning capacity of mice tested by water maze experiment,number of neuronal nissl body and neuronal morphology in brain tissue as observed by nissl staining,and the concentration of GSH in the serum of mice was measured by a kit.Expression of genes related to the Nrf2/Keap1 signalling pathway and apoptotic pathways were examined using realtime fluorescence quantitative PCR.Ginsenoside CK improves the learning and memory ability of cognitively impaired mice,inhibits hippocampal neuronal damage in mouse brain tissue,increases the level of GSH in serum,regulates the expression of genes related to Nrf2/Keap1 signaling pathway in brain,and inhibits apoptosis in brain tissue.Ginsenoside CK can inhibit oxidative damage and apoptosis of neurons in mouse brain tissue and improve cognitive impairment in mice by regulating genes related to the Nrf2/Keap1 signaling pathway.
