摘  要:马乳营养成分最接近人乳。研究表明,马乳对人体免疫功能有调节作用,对癌症有一定的预防作用。文章中观察到1种全新马乳源性多肽,序列为VAPFPQPVVPYPQR,对非小细胞肺癌有一定抑制作用,但其确切机制尚不清楚。文章旨在研究该肽对非小细胞肺癌的抑制机制,重点关注线粒体动力学和细胞周期。在培养肺癌细胞A549时用该肽进行干预,观察其对细胞凋亡、细胞增殖和细胞周期的影响。检测发现,MFN 1和DRP 1蛋白被下调,MFN 1和DRP 1的下调抑制了线粒体的融合和分裂,导致线粒体延长。SIRT2-SKP2-CDK1通路也受到抑制,CDK 1蛋白及SIRT2均呈下调趋势,导致细胞周期在G1期受到抑制。Horse milk is the closest emulsion to human milk in terms of nutritional content.Studies have shown that horse milk has a regulatory effect on human immune function and has a preventive effect on cancer.We observed a novel horse milk-derived peptide with the sequence VAPFPQPVVPYPQR,which has a certain inhibitory effect on non-small cell lung cancer,but the exact mechanism is unknown.The aim of this study was to investigate the inhibitory mechanism of this peptide on non-small cell lung cancer,focusing on both mitochondrial dynamics and cell cycle.The peptide was administered in the culture of lung cancer cells A549,and its effects on apoptosis,cell proliferation and cell cycle were observed.MFN 1 and DRP 1 proteins were detected and found to be down-regulated,and the down-regulation of MFN1 and DRP1 inhibited mitochondrial fusion and fission,resulting in the prolongation of mitochondria.The SIRT2-SKP2-CDK1 pathway was also inhibited,resulting in cell cycle inhibition in G1 phase.
