摘  要:文章开发一种基于重组酶介导等温核酸扩增技术(Recombinase Aided Amplification,RAA)的方法,实现对食品中骆驼源性成分快速检测。针对骆驼细胞色素B(Cytochrome b,Cyt b)基因设计特异性引物,对建立的RAA体系优化其反应扩增条件,评估其特异性和灵敏度,并用建立的RAA法与GB/T 38164标准方法分别对24份市售骆驼制品进行检测并比对结果。结果表明,本研究建立的RAA法特异性强,乳及乳制品检出限为1.0%,肉及肉制品检出限为0.1%,且2种方法对24份骆驼乳肉及其制品检测结果一致。因此,本研究开发的RAA方法具有快速、特异性强、灵敏度高、操作简单等优点,市场应用前景广阔。Developing a method based on Recombinase Aided Amplification(RAA)for the rapid detection of camel-derived ingredients in food.Specific primers were designed targeting the Cytochrome b(Cyt b)gene of camels,and the RAA system's reaction conditions were optimized.The specificity and sensitivity of the established RAA method were evaluated.Twenty-four commercially available camel products were tested using both the established RAA method and the GB/T 38164 standard method,and the results were compared.The outcomes revealed that the developed RAA method exhibited strong specificity,with detection limits of 0.1%for meat and meat products and 1.0%for milk and dairy products.The results from both methods were consistent for all 24 camel products.Therefore,the RAA method developed in this study possesses advantages such as rapidity,strong specificity,high sensitivity,and operational simplicity,suggesting broad applications in the field.
