摘 要:目的建立鸡头黄精质量评价方法,比较不同种植方式鸡头黄精的质量差异,为鸡头黄精种植方式的优选提供参考。方法采用高效液相色谱-蒸发光散射检测法(HPLC-ELSD法)建立鸡头黄精中果糖、葡萄糖、蔗糖含量测定方法;采用高效液相色谱-二极管阵列检测法(HPLC-DAD法)建立鸡头黄精中亚油酸、5-羟基麦芽酚、5-羟甲基糠醛、黄精碱A含量测定方法;采用紫外-可见分光光度法测定鸡头黄精中多糖含量。结果所测定不同种植方式鸡头黄精中多糖、果糖、亚油酸、5-羟基麦芽酚,以林下种植方式所得药材含量最高,蔗糖以露天种植方式所得药材含量最高,5-羟甲基糠醛、黄精碱A则以与玉米进行套种所得药材含量最高;OPLS-DA分析结果表明,所有鸡头黄精药材以种植方式不同聚为不同的类群。结论所建立鸡头黄精质量评价方法稳定可行,不同种植方式鸡头黄精质量差异明显,以林下种植模式下药材成分累计含量最高。Objective To develop a quality evaluation method of Polygonatum sibiricum Red.medicinal materials and compare the quality differences of Polygonatum sibiricum Red.medicinal materials obtained by different planting patterns,so as to provide reference for the optimization of planting pattern of Polygonatum sibiricum Red.Methods HPLC-ELSD was used for the determination of fructose,glucose and sucrose,and HPLC-DAD was used for the determination of linoleic acid,5-hydroxymaltol,5-hydroxymethylfurfural and polygonatine A in Polygonatum sibiricum Red.medicinal materials.Ultraviolet-visible spectrophotometry was used for the determination of polysaccharide in Polygonatum sibiricum Red.medicinal materials.Results The contents of polysaccharide,fructose,linoleic acid,5-hydroxymaltol in Polygonatum sibiricum Red.medicinal materials obtained by different planting patterns were determined,which were the highest in the medicinal materials obtained by understory planting.The content of sucrose in the medicinal materials obtained by open planting was the highest.The contents of 5-hydroxymethylfurfural and polygonatine A were the highest in the medicinal materials obtained by interplanting with maize.The results of OPLS-DA analysis showed that all Polygonatum sibiricum Red.medicinal materials were clustered into different groups based on different planting patterns.Conclusion The quality evaluation method of Polygonatum sibiricum Red.medicinal materials is stable and feasible.The quality of Polygonatum sibiricum Red.medicinal materials is of significant difference obtained by different planting patterns.The cumulative content of the effective components is the highest in the medicinal materials obtained by understory planting.
