摘 要:外泌体是一种由多种细胞分泌的、直径30~200 nm,具有与细胞相同的脂质双分子层结构,富含特定蛋白质、脂质、核酸及各种代谢物的细胞外囊泡,起源于质膜形成的早期内体,在细胞间信息传递过程中起重要作用。外泌体能够进入细胞并释放其内含物,可作为药物递送的理想载体。目前优选的药物递送系统载体都有其缺陷,而外泌体因其自身特性在发展药物递送系统方面有重要潜能,有望弥补现有药物递送系统载体的不足。本文就外泌体药物递送系统的构建及其在癌症、精神疾病及病毒性疾病治疗等方面应用的最新进展展开综述。Exosomes are extracellular vesicles secreted by various cells,with a diameter of 30-200 nm,which have the same lipid bilayer membrane structure as cells and are rich in specific proteins,lipids,nucleic acids,and various metabolites.Originating from early endosomes formed by the plasma membrane,exosomes play a crucial role in intercellular information transmission.Exosomes can enter cells and release their contents,making them ideal carriers for drug delivery.At present,the preferred drug delivery system carriers all have some defects,and exosomes have important potential in the development of drug delivery systems due to their own characteristics,which is expected to make up for the shortcomings of the existing drug delivery system carriers.This review summarizes the latest advances in the construction of exosome-based drug delivery systems and their applications in the treatment of cancer,mental disorders and viral diseases.
