摘 要:为研究不同短日照诱导对小豆叶片代谢物质的影响。本研究以晚熟品种‘冀红16’为材料,设置8 h光/16 h暗和10 h光/14 h暗两种短日照诱导并处理14 d(简称:8 h14 d和10 h14 d),利用超高效液相色谱串联质谱(UPLC-MS/MS)技术对8 h14 d vs 10 h14 d比较组进行代谢定性研究。结果表明:随着日照时间缩短,差异代谢物质不断增多,8 h14 d vs 10 h14 d比较组共获得10大类58种代谢物质。10大类代谢物质分别是黄酮、酚酸类、木脂素和香豆素、脂质、氨基酸及其衍生物、生物碱、萜类、有机酸、核苷酸及其衍生物、醌类。58种代谢物质中48种上调代谢物质,10种下调代谢物质。差异代谢物的动态变化表明,10种上调代谢物质属于黄酮、木脂素和香豆素类,10种下调代谢物质属于酚酸、脂质、氨基酸及其衍生物、萜类、有机酸类。KEGG富集结果显示,代谢物质较多,占比较高的前5条代谢通路分别是代谢途径、次生代谢生物合成、黄酮、异黄酮、氨基酸生物合成途径,差异代谢物数量(占比)分别为10(76.92%)、9(69.23%)、3(23.08%)、3(23.08%)、3(23.08%),其中,富集差异显著(P<0.5)的3条代谢通路分别是次生代谢生物合成、黄酮、异黄酮代谢通路,上调代谢物的数量分别为6、3、3,下调代谢物的数量分别为3、0、0,次生代谢物途径中上调代谢物质多为酮类,下调代谢物质多为酸类。以上研究结果表明,小豆叶片通过上调酮类代谢物质数量和下调酸类代谢物质数量来响应短日照诱导,而8 h14 d严重影响小豆植株的碳氮代谢,诱导植株酸化,抑制植株生长,认为10 h14 d是合理的短日照诱导时间。To study the effects of different short-day photoperiod on metabolic substances in adzuki bean leaves,the late-maturity variety‘Ji hong 16’was used as the material in this study,and seedlings were grown for 14 d under two short-day photoperiod conditions of 8 h light/16 h dark and 10 h light/14 h dark(abbreviated:8 h14 d and 10 h14 d).The Ultra-high Performance Liquid Chromatography by Tandem Mass Spectrometry(UPLC-M S/MS)technology was applied to compare metabolic difference between treatment 8 h14 d and 10 h14 d.The results demonstrated that with the shortening of photoperiod,the differential metabolic substances were continuously increased,and 58 metabolic substances were obtained in the 8 h14 d vs 10 h14 d comparison group.The 10 major classes of the differential metabolic substances were flavonoids,phenolic acids,lignans and coumarins,lipids,amino acids and derivatives,alkaloids,terpenoids,organic acids,nucleotides and derivatives,quinones.There were 58 differential metabolic substances,including 48 up-regulated and 10 down-regulated metabolic substances.10 up-regulated metabolites belonged to flavonoids,lignans,and coumarins,and 10 down-regulated metabolites belonged to phenolic acids,lipids,amino acids and their derivatives,terpenoids,organic acids.KEGG enrichment results showed that the top 5 metabolic pathways were metabolic pathway,secondary metabolic biosynthesis,flavonoids,isoflavone,amino acid biosynthesis pathway,in which the number of differential metabolites(proportion)were 10(76.92%),9(69.23%),3(23.08%),3(23.08%),3(23.08%),respectively.Among them,secondary metabolic biosynthesis,flavone and isoflavone three metabolism pathways were enriched significantly(P<0.5).The number of up-regulated metabolites were 6,3,3,and down-regulated metabolites were 3,0,0,respectively,while up-regulated metabolites were mostly ketones and down-regulated metabolites were mostly acids in secondary metabolite pathways.The above results suggested that adzuki bean leaves responded to short-day photoperiod by up-regula
