生防菌Ba1-7对大豆胞囊线虫的防效及对大豆根际微生物群落的影响

摘  要:为明确阿氏芽胞杆菌(Bacillus aryabhattai) Ba1-7对大豆胞囊线虫病的防治效果以及对根际土壤微生物的影响,本研究采用室内盆栽试验和大田试验探究该菌株对大豆胞囊线虫病的防治效果,利用高通量测序技术,采用Krona工具对物种注释结果进行可视化分析,明确田间大豆胞囊线虫胁迫下Bal-7对大豆根际土壤微生物群落结构和组成的影响。结果表明:生防菌Ba1-7处理的大豆株高、主根长以及产量等显著高于对照处理,产量比对照处理增加29%。对大豆胞囊线虫的盆栽和田间防效分别为63.35%和67.28%,说明该菌株对大豆有促生长作用以及对大豆胞囊线虫有防治效果。高通量测序及分析表明两组处理的土壤样本中的细菌菌群归属于35个门,112个纲,263个目,435个科,793个属;真菌菌群归属于12个门,36个纲,81个目,175个科,325个属。生防菌Ba1-7处理提高了土壤中微生物的群落多样性,降低了细菌和真菌的物种总数和群落丰富度,但对微生物群落多样性结构和组成无显著差异,变形菌门、放线菌门、绿弯菌门和子囊菌门以及假单杆菌属和单杆菌属丰度有所增加,说明该菌株可能是通过影响微生物群落的菌群丰度来对大豆胞囊线虫进行防治,对土壤生态环境是安全可靠的。To clarify the control effect of Bacillus aryabhattai Ba1-7 on soybean cyst nematode disease and its impact on rhizosphere soil microorganisms,this study used indoor pot experiments and field experiments to explore the control effect of this strain on soybean cyst nematode disease.Using high-throughput sequencing technology,Krona tools were used to visualize the results of species annotation,and the effects of Bal-7 on the microbial community structure and composition of soybean rhizosphere soil under soybean cyst nematode stress in the field were analyzed.The results showed that the plant height,main root length,and yield of soybean treated with the biocontrol bacterium Ba1-7 were significantly higher than the control treatment,the yield was increased by 29%compared to the control treatment.The potted and field control effects on soybean cyst nematode were 63.33%and 67.28%,respectively,indicating that the strain has a growth promoting effect on soybeans and a control effect on soybean cyst nematode.After a growing season,high-throughput sequencing results showed that the treatment with biocontrol bacteria Ba1-7 increased the diversity of microbial communities in the soil,reduced the total number and richness of bacterial and fungal species,but did not show significant differences in the structure and composition of microbial community diversity.However,the abundance of Proteobacteria,Actinobacteria,Chloroflexia,p_Ascomycota,Pseudomonas,Pseudarthrobacter and Solirubrobacter increased,indicating that this strain may have controlled soybean cyst nematodes by affecting the abundance of microbial communities,which is safe and reliable for the soil ecological environment.
