摘 要:为验证藻类智能检测仪的可靠性,以及结合洱海流域的实际情况,探讨藻类智能检测仪在洱海流域的适用性,于2021年11月—2022年11月,对洱海流域的藻类开展藻类智能检测仪和人工检测比对实验。结果表明:藻类智能检测仪与人工比对的相对偏差范围在-78.91%~96.50%,重复实验的相对偏差绝对值范围在0%~96.07%,样品浓缩5倍时的方法检出限为0.2×10^(4)个/L,藻细胞密度在1000~10000万个/L,藻类智能检测仪与人工比对的一致性较好。藻类智能检测仪与人工检测藻细胞密度变化趋势总体一致,在一定程度上可以反映藻类的变化情况,在种属组成和优势种鉴定存在一定差异,也基本能反映藻类种群结构情况。鉴于洱海流域的特点,藻类多样性较高,种群结构随季节、时间变化较快,藻类智能检测仪需要进一步相对准确地定量藻细胞密度和优势种,图谱库需进行更新补充、训练、维护优化,检测过程应经常更换卡夹,确保数据的准确性。To verify the reliability of an intelligent algae detector and explore its applicability in the Erhai Lake Basin based on actual situations of the Erhai Lake Basin,a comparative experiment between the intelligent algae detector and manual detection was conducted from November 2021 to November 2022 for the algae in the Erhai Lake Basin.The results showed that The relative deviation range between the intelligent algae detector and manual detection was-78.91%to 96.50%,and the absolute value range of relative deviation in repeated experiments was 0.00%to 96.07%.When the samples were concentrated for five times,the method detection limit was 0.2×104 cells/L.When the algal cell density ranged from 10 million to 100 million cells/L,the intelligent algae detector demonstrated good consistency with manual detection.Generally,the trends in algal cell density measured with the intelligent algae detector were consistent with those obtained through manual detection,indicating that the intelligent algae detector can reflect changes in algae to a specific extent.However,although there were some differences in species composition and identification of dominant species,the detector was still able to indicate basic algal population structure.In view of the characteristics of the Erhai Lake Basin,high algal diversity and rapid changes in population structure depending on season and time,the intelligent algae detector needs further improvement to accurately quantify algal cell density and identify dominant species.The spectral library also requires regular updates,training,and maintenance optimization.Additionally,it is recommended to frequently replace the card clips during the detection process to ensure data accuracy.
