摘　　要：目的旨在探讨基于PI3K/Akt信号通路探讨胎宁方在复发性流产中的作用机制。方法将80只8周龄SPF级CBA/J雌性大鼠随机分为对照组、模型组、胎宁方低剂量组、胎宁方中剂量组、胎宁方高剂量组,采用合笼交配发情的DBA/2雄鼠和CBA/J雌鼠(1∶2),制备复发性流产模型;合笼交配发情的BALB/c雄性大鼠和CBA/J雌鼠(1∶2),制备正常的妊娠模型。造模成功后胎宁方各处理组分别给予低、中、高剂量的胎宁方灌胃14 d,检测大鼠子宫脏器指数、胚胎丢失率、流产率及蜕膜组织中相关指标的表达情况,采用酶联免疫吸附法测定HCG、t-PA、APC、AT-Ⅲ水平,Western blot法及qPCR法检测PI3K/Akt信号通路的蛋白表达及mRNA水平。结果与对照组相比,模型组子宫脏器指数、胚胎丢失率及流产率均较高(P<0.05),提示造模成功。与模型组相比,不同剂量的胎宁方组大鼠子宫脏器指数、胚胎丢失率及流产率均显著升高,并且具有一定的剂量效应,但仍低于对照组(P<0.05)。对照组大鼠蜕膜组织细胞血管丰富,界限分明,排列有规律;模型组大鼠蜕膜组织细胞血管及间质血管数量减少,间质水肿,存在炎性细胞,部分蜕膜细胞坏死,排列紊乱,子宫内膜蜕膜样变;不同剂量的胎宁方组大鼠蜕膜组织细胞血管数量增多,间质水肿减少,细胞排列规律,呈对照组变化,且蜕膜组织细胞形态改变情况与剂量呈正相关。与对照组相比,模型组HCG、t-PA、APC、AT-Ⅲ均较高(P<0.05)。与模型组相比,不同剂量的胎宁方组大鼠HCG、t-PA、APC、AT-Ⅲ均显著升高,并且具有一定的剂量效应,但仍低于对照组(P<0.05)。与对照组相比,模型组PI3K、p-Akt蛋白均较高(P<0.05)。与模型组相比,不同剂量的胎宁方组大鼠PI3K、p-Akt蛋白均显著升高,并且具有一定的剂量效应,但仍低于对照组(P<0.05)。与对照组相比,模型组PI3K、p-AktmRNA均较高(P<0.05)。与模型组相比,不同剂量�Objective This study was to investigate the mechanism of Taining Prescription(胎宁方)in recurrent abortion based on phosphatidylinositol 3-kinase(PI3K)/phosphate kinase B(Akt)signaling pathway.Methods Eighty 8-week-old SPF CBA/J female rats were randomly divided into control group,model group,Taining Prescriptionlow dose group,Taining Prescription medium dose group and Taining Prescription high dose group.The recurrent abortion model was prepared by using cage mating estrus DBA/2 male rats and CBA/J female rats(1∶2).BALB/c male rats and CBA/J female rats(1∶2)in cage mating estrus were used to create a normal pregnancy model.After successful modeling,each treatment group was given the low,medium and high doses of Taining Prescription for 14 days,respectively,to detect the uterine organ index,embryo loss rate,abortion rate and the expressions of related indicators in decidua tissue of rats,and the levels of human chorionic gonadotropin(HCG),tissue-type plasminogenactivator(t-PA),antigen presenting cell(APC)and antithrombin III(AT-Ⅲ)were determined by enzyme-linked immunosorbent assay.The protein expressions and mRNA levels of PI3K/Akt signaling pathway were detected by Western blot and qPCR.Results Compared with those in the control group,the uterine organ index,embryo loss rate and abortion rate in the model group were higher(P<0.05),indicating successful modeling.Compared with those of the model group,the uterine organ index,embryo loss rate and abortion rate of rats in different dosegroups of Taining Prescription were significantly increased,and there was a certain dose effect,but they were still lower than those of the control group(P<0.05).The blood vessels in the decidual tissue cells of the control groupwere abundant,clearly definedand arranged in a regular pattern.The model group showed a decrease in the number of blood vessels and interstitial blood vessels in the decidual tissue cells,interstitial edema,the presence of inflammatory cells,partial necrosis and disordered arrangement of decidual cel
