淫羊藿苷和朝藿定C对前列腺癌裸鼠模型肿瘤细胞抑制作用的实验研究

摘　　要：目的探讨淫羊藿苷和朝藿定C对前列腺癌裸鼠模型肿瘤细胞抑制作用的实验。方法清洁级雄性SCID小鼠48只,雄性,采用前列腺腺体背外侧包膜内注射人前列腺癌细胞株(human prostate cancer cell line,LNCaP)悬液的方法构建前列腺癌原位移植瘤模型,采用随机数字表随机均分为模型组、雷帕霉素组、淫羊藿苷组、朝藿定C组,每组12只。于建模2周后,淫羊藿苷组予以10 mg·kg^(-1)淫羊藿苷,朝藿定C予以10 mg·kg^(-1)朝藿定C,模型组给予等容量生理盐水灌胃给药,每日1次,连续60 d。4组于治疗前和治疗结束时各取6只小鼠肿瘤细胞生长情况评估,ELISA法检测白细胞介素6(interleukin-6,IL-6)、白细胞介素8(interleukin-8,IL-8)、转化生长因子β(trans forming growth factor-β,TGF-β)、前列腺特异性抗原(prostate specific antigen,PSA)的表达水平,FCM检测组织Treg的表达,western blotting检测caspase-3、bcl-2的表达。应用SPSS 23.0软件进行分析。结果与治疗前相比,治疗后雷帕霉素组、淫羊藿苷组、朝藿定C组瘤质量、瘤体积降低,抑瘤率增加(P<0.05);模型组治疗前后瘤质量、瘤体积、抑瘤率相比差异无统计学意义(P>0.05)。治疗后与模型组相比,雷帕霉素组、淫羊藿苷组、朝藿定C组瘤质量、瘤体积较低,抑瘤率较高(P<0.05);与雷帕霉素组相比,朝藿定C组瘤质量、瘤体积较高,抑瘤率较低(P<0.05);淫羊藿苷组与雷帕霉素组瘤质量、瘤体积、抑瘤率相比差异无统计学意义(P>0.05)。与模型组相比,雷帕霉素组、淫羊藿苷组、朝藿定C组血清IL-6、IL-8、TGF-β水平较低(P<0.05);与雷帕霉素组相比,朝藿定C组血清IL-6、IL-8、TGF-β水平较高(P<0.05),淫羊藿苷组与雷帕霉素组血清IL-6、IL-8、TGF-β水平相比差异无统计学意义(P>0.05)。与模型组相比,雷帕霉素组、淫羊藿苷组、朝藿定C组血清PSA水平、血液细胞Treg数量较低(P<0.05);与雷帕霉素组相比,朝藿定C组血�Objective To investigate the inhibitory effects of icariin and chaohodin C on tumor cells in a nude mouse model of prostate cancer.Methods Forty-eight male SCID mice of clean grade were injected with human prostate cancer cell line(LNCaP)suspension into the dorsal lateral capsule of the prostate gland to construct the orthotopic tumor model of prostate cancer,and were randomly divided into model group,Sirolimus group,icariin group and chaohodine C group,with 12 in each group.Two weeks after modeling,the icariin group was given icariin(10 mg·kg^(-1)),while the icariin C group was given chaohodine C(10 mg·kg^(-1)).The model group was given an equal volume of physiological saline by gavage once a day for 60 consecutive days.Six mice were taken from each of the four groups before and after treatment to evaluate the growth of tumor cells.ELISA was used to detect the levels of interleukin-6(IL-6),interleukin-8(IL-8),transforming growth factor-β(TGF-β)and prostate specific antigen(PSA).FCM was used to detect the expression level of Treg expression in tissues,and Western blotting detected the expressions of caspase-3 and bcl-2.SPSS 23.0 software was applied for analysis.Results Compared with those before treatment,the tumor mass and volume of Sirolimus group,icariin group and chaohodine C group decreased after treatment,and the tumor inhibition rate increased(P<0.05).There was no statistically significant difference in tumor mass,tumor volume or tumor inhibition rate in the model group before and after treatment(P>0.05).After treatment,compared with those of the model group,the tumor mass and volume of Sirolimus group,icariin group and chaohodine C group were lower,and the tumor inhibition rate was higher(P<0.05).Compared with those of the Sirolimus group,the tumor mass and volume of group C were higher,and the tumor inhibition rate was lower(P<0.05).There was no significant difference in tumor mass,tumor volume or tumor inhibition rate between icariin group and Sirolimus group(P>0.05).Compared with those of the mode
