中药复方基于Wnt信号通路探讨类风湿关节炎与肿瘤的相关性

摘　　要：Wnt信号通路是参与细胞分化、肢体发育、关节形成、癌变和干细胞维持等多种生物学现象的细胞因子,在滑膜增生、骨形成、骨吸收和肿瘤发生中发挥作用。升高的Wnt信号也通过多种机制,包括维持癌症干细胞群、增强DNA损伤修复、促进转录可塑性和促进免疫逃避等对多种传统和靶向癌症治疗产生抗性。近年来,中药针对Wnt信号通路调控及其相关配体、蛋白和关键酶的表达来探索类风湿关节炎(rheumatoid arthritis,RA)的作用机制也被大量报道。该文基于Wnt信号通路,通过综述近几年来中药复方调控Wnt治疗肿瘤和RA的研究成果,探讨RA与肿瘤发生机制的相关性,进一步为临床运用中药复方治疗患肿瘤的类风湿关节炎患者及其及抑制剂的开发提供思路。The Wnt signaling pathway is a cytokine involved in various biological phenomena such as cell differentiation,limb development,joint formation,carcinogenesis and stem cell maintenance,and plays a role in synovial hyperplasia,bone formation,bone resorption and tumorigenesis.Elevated Wnt signaling also contributes to resistance to a variety of conventional and targeted cancer therapies through multiple mechanisms,including maintenance of cancer stem cell populations,enhanced DNA damage repair,promotion of transcriptional plasticity and promotion of immune evasion.In recent years,the mechanism of action of traditional Chinese medicine on the regulation of Wnt signaling pathway and the expressions of related ligands,proteins and key enzymes to explore rheumatoid arthritis(RA)has also been extensively reported.Based on the Wnt signaling pathway,this paper reviewed the research results of traditional Chinese medicine regulating Wnt in the treatment of tumors and RA in recent years,discussed the similarity of the mechanism of arthritis and tumorigenesis,and further provided the clinical application of traditional Chinese medicine for the treatment of arthritis patients with tumors and the development of their inhibitors.
