摘  要:目的:观察温阳补肾灸对APP/PS1双转基因小鼠突触可塑性损伤及Aβ的影响,探讨温阳补肾灸治疗阿尔茨海默病(AD)的作用机制。方法:10只C57BL/6J小鼠作为正常组;20只APP/PS1双转基因小鼠随机分为模型组和艾灸组,各10只。艾灸组悬灸“百会穴”“命门穴”,隔龟板灸“关元穴”,每次30 min,1次/d,共3周。Morris水迷宫实验检测干预前后小鼠学习记忆能力;Western blot检测线粒体分裂蛋白1(Fis1)、线粒体融合蛋白1(OPA1)、突触后致密物-95(PSD-95)和突触素(SYN)的蛋白表达;透射电镜观察突触形态结构;免疫组织化学法检测β-淀粉样蛋白_(1-42)(Aβ_(1-42))蛋白表达。结果:干预前,模型组与正常组比较逃避潜伏期时间延长,差异具有统计学意义(P<0.05);模型组与正常组比较,Fis1和Aβ_(1-42)蛋白表达均升高,差异具有统计学意义(P<0.05),OPA1、PSD-95和SYN蛋白表达均下降,差异具有统计学意义(P<0.05)。电镜下观察模型组的突触超微结构损伤,突触间隙增宽。干预后,艾灸组与模型组比较逃避潜伏期时间缩短,差异具有统计学意义(P<0.05);Fis1和Aβ_(1-42)蛋白均表达下降,差异具有统计学意义(P<0.05);OPA1、PSD-95和SYN蛋白表达均升高,差异具有统计学意义(P<0.05);电镜下观察艾灸组突触形态结构改善,突触间隙宽度减小。结论:温阳补肾灸通过抑制Aβ沉积减少线粒体损伤,改善突触可塑性损伤,进而改善认知功能。Objective:To observe the effect of Wenyang Bushen(warming Yang and tonifying Kidney)moxibustion on synaptic plasticity damage and Aβin APP/PS1 double transgenic mice,thus to explore the mechanism of Wenyang Bushen moxibustion in treatment of Alzheimer's disease(AD).Methods:10 C57BL/6J mice were used as the normal control.20 APP/PS1 mice were randomly divided into the model group and the moxibustion group,with 10 mice in each group.The moxibustion group used suspension moxibustion on Baihui(DU20)and Mingmen(DU04),and turtle plate moxibustion on Guanyuan(RN4)for 30 minutes,once per day for three weeks.The learning-memory ability was detected by Morris water maze tests in the mice.The protein expressions of Fis1,OPA1,PSD-95 and SYN were detected by Western blot.Morphological structure of synapses was observed by transmission electron microscopy.The protein expression of Aβ_(1-42)was detected by immunohistochemistry method.Results:The escape latency time of the model group was prolonged compared to the normal group(P<0.05).Compared with those in the normal group,the protein expressions of Fis1 and Aβ_(1-42)were increased(P<0.05),and the protein expressions of OPA1,PSD-95 and SYN were decreased in the model group(P<0.05).Under electron microscopy,the model group showed synaptic ultrastructural damage,including widened synaptic clefts.After the intervention,the moxibustion group demonstrated a shortened escape latency compared to the model group(P<0.05).The protein expressions of Fis1 and Aβ_(1-42)were decreased(P<0.05),and the protein expressions of OPA1,PSD-95,and SYN were increased in the moxibustion group(P<0.05).Under electron microscopy,the moxibustion group exhibited improved synaptic morphology,with a reduced synaptic cleft width.Conclusion:Wenyang Bushen moxibustion reduces mitochondrial damage by inhibiting Aβ_(1-42)deposition,which in turn improves synaptic plasticity impairment and enhances cognitive function.
