摘 要:为了探讨光肩星天牛雌虫复眼基因对光信号的反应,并挖掘与生物防治相关的基因,为光肩星天牛合理防控提供理论基础。利用比较转录组学方法,在光照和黑暗条件下进行4个处理差异基因表达分析,分别为光照腹部(LV)、光照背部(LD)、黑暗腹部(DV)、黑暗背部(DD)的差异基因表达分析。结果表明:(1)共获得16200个基因,其中差异基因4166个,DV_VS_DD、LV_VS_LD、LV_VS_DV、LV_VS_DD中差异基因分别为649、2588、1693、3522个。(2)GO功能分析显示差异基因主要富集在肽生物合成过程、翻译、酰胺生物合成过程、肽代谢过程、细胞大分子生物合成过程。(3)KEGG富集分析显示差异基因主要集中在核糖体、氧化磷酸化、热生成作用等KEGG途径。(4)与生物防治相关基因有106个,其中与嗅觉相关的31个,蜕皮相关的58个,几丁质合成相关的6个和木质纤维素相关的11个。该结果为RNA干扰靶标基因防治光肩星天牛提供了理论基础。The research aims to investigate the gene response of Anoplophora glabripennis compound eyes to light signals and explore the genes related to biological control of Anoplophora glabripennis,providing theoretical basis for the control of this beetle.Comparative transcriptome analysis method was used to analyze the differential gene expression of 4 treatments:light tretment to ventral area(LV),light tretment to dorsal area(LD),dark treatment to ventral area(DV)and dark treatment to dorsal area(DD).The results showed that:(1)Transcriptome sequencing was performed on 12 samples,and 16200 genes were obtained;A total of 4166 differential expression genes were found,including 649,2588,1693 and 3522 d in DV vs DD,LV_vs_LD,LV_vs_DV,and LV_vs_DD,respectively.(2)GO term analysis showed that differential genes were mainly enriched in peptide biosynthesis,translation,amide biosynthesis,peptide metabolism,and cellular macromolecular biosynthesis.(3)KEGG pathway analysis showed that differential genes were mainly concentrated in ribosome,oxidative phosphorylation and thermogenesis.(4)Furthermore,106 genes related to biological control were found,including 31 olfaction-related genes,58 molting-related genes,6 chitin synthesis-related genes and 11 lignocellulose-related genes.The result provides a theoretical basis for using RNA interference to control A.glabripennis.
