摘 要:为明确白榆种质资源的亲缘关系,了解种质的遗传基础,为资源保护和品种选育提供依据,采用SSR分子标记对河北省邓栅子林场白榆国家林木种质资源库中的83份白榆种质资源进行了遗传多样性与聚类分析。结果表明:(1)11对引物共扩增出59条多态性条带,多态性比率达98.33%。平均多态性信息含量(PIC)为0.6980。(2)平均等位基因数(Na)为1.9833,平均有效等位基因数(Ne)为1.3011,平均Nei’s基因多样性指数(H)为0.2040,平均Shannon多样性指数(I)为0.3387,白榆种质资源在DNA水平上存在丰富的遗传变异。(3)群体间的遗传分化系数(Gst)为0.1238,白榆群体内部遗传变异显著高于群体间变异。遗传距离和遗传相似性分析发现,参试3个群体的亲缘关系较近,遗传背景相似。(4)83份白榆种质平均遗传距离为0.6201,经UPMGA聚类分析,可将白榆种质分为6个类群,聚类结果分析和主成分分析结果一致性较强。研究发现邓栅子林场来自不同地区的白榆种质资源遗传多样性较为丰富,亲缘关系较近。The research aims to clarify the genetic relationship of Ulmus pumila germplasm resources,understand the genetic basis of germplasm,and provide a basis for resource protection and variety selection.SSR molecular marker method was adopted to conduct genetic diversity and cluster analysis on 83 U.pumila germplasm resources in the National Forest Germplasm Database of Deng Zhazi Forest Farm in Hebei Province.The results showed that:(1)A total of 59 polymorphic bands were amplified by 11 pairs of primers,with a polymorphism rate of 98.33%.The average polymorphism information content(PIC)was 0.6980.(2)The average number of alleles(Na)was 1.9833,the average number of effective alleles(Ne)was 1.3011,the average Nei's gene diversity index(H)was 0.2040,and the average Shannon diversity index(I)was 0.3387.The U.pumila germplasm resources had rich genetic variation at the DNA level.(3)The genetic differentiation coefficient(Gst)between populations was 0.1238,and the genetic variation within the U.pumila population was significantly higher than that between populations.The analysis of genetic distance and genetic similarity showed that the three populations were closely related and had similar genetic backgrounds.(4)The average genetic distance of 83 U.pumila germplasms was 0.6201.UPMGA cluster analysis showed that U.pumila germplasms could be divided into 6 groups,and the results of cluster analysis and principal component analysis were consistent.This study found that the genetic diversity of U.pumila germplasm resources from different regions in Deng Zhazi Forest Farm was relatively rich and there were close genetic relationships among those diversities.
