摘 要:眼部结构的复杂性使普通眼用制剂的生物利用度低、患者依从性差。缓释长效药物递送系统因其可以克服眼部屏障或者在眼内形成药物储库,提高药物生物利用度,减少患者用药频次,在治疗多种眼部慢性疾病中具有显著的临床优势。根据剂型分类,对已上市的缓释长效眼用制剂、处于临床研究中以及最近文献报道的此类制剂的缓释作用和原理进行综述,以期为眼部缓释长效制剂的研发提供新思路。The common ophthalmic preparations have limitations including low drug bioavailability and poor patient compliance due to the complexity of the eye structure.Sustained-release and long-acting drug delivery systems have significant clinical advantages in the treatment of a variety of ocular chronic diseases,as they can overcome the eye barriers or form a drug reservoir in the eye,thereby improving drug bioavailability and reducing patient medication frequency.According to the classification of dosage forms,this review mainly introduces sustained-release and long-acting ophthalmic preparations commercially available,along with those in current clinical research and recent reports in literature.The sustained-release mechanism and principle of these preparations are described,so as to provide new insights for the development of sustained-release and long-acting ophthalmic preparations.
