摘　　要：目的探讨去甲斑蝥素(norcantharidine,NCTD)对肝细胞生长因子(hepatocyte growth factor,HGF)诱导的下咽癌FaDu细胞(简称FaDu细胞)侵袭和迁移的影响及机制。方法采用体外培养FaDu细胞,HGF诱导下咽癌细胞侵袭和迁移,以不同浓度NCTD处理FaDu细胞。运用Transwell法检测NCTD对FaDu细胞侵袭和迁移的影响,Western blot检测基质金属蛋白酶9(matrix metalloproteinase-9,MMP-9)和尿激酶型纤溶酶原激活剂(u-PA)蛋白表达的变化。结果NCTD可以显著降低FaDu细胞的相对生存率,并且其抑制效应呈现出剂量依赖性。Trasnwell侵袭实验和迁移试验中,各NCTD处理组穿过Transwell小室细胞数与HGF处理组相比明显减少,差异均有统计学意义(P均<0.01),随NCTD浓度的升高,侵袭和迁移到下层小室的细胞数量逐渐减少。Western blot实验结果显示NCTD可抑制HGF诱导的FaDu细胞中MMP-9和u-PA蛋白的表达(P<0.01),呈剂量依赖型。结论NCTD具有抑制HGF诱导的FaDu细胞侵袭和迁移的作用,其机制可能与抑制MMP-9、u-PA蛋白表达有关。Objective To investigate effect of Norcantharidin on HGF-induced invasion and metastasis of human hypopharyngeal carcinoma cells FaDu.Methods Human hypopharyngeal carcinoma cells,FaDu was cultuerd in vitro,invasion and migration were induced by HGF and different concentrations of NCTD were added.Transwell method was adopted to measure the effect of NCTD on the invasion and metastasis of human hypopharyngeal cancer cells in different groups.Western blot method was used to measure the expressions of u-PA and MMP-9 protein.Results Norcantharidin inhibited the invasion and metastasis of FaDu cells in a concentration-dependent manner.The results of Transwell assay showed that norcantharidin can inhibit FaDu cells invasion capability at different concentration(P<0.01)and the inhibitory effect of Norcantharidin increased along with the increase of concentration.MMP-9 and u-PA protein levels expression were significantly decreased after treatment of the FaDu cells with NCTD,the inhibitory effect of Norcantharidin in a concentration-dependent manner.Conclusions Norcantharidin can inhibit hypopharyngeal carcinoma cell invasion and metastasis,which might be related to that NCTD could regulate the expressions of MMP-9 and u-PA expression.
