网络药理学与生物信息学联合分析烟酸在骨性关节炎中的潜在治疗基因靶点

摘　　要：目的利用GEO数据库联合网络药理学讨论烟酸治疗骨性关节炎的可能作用机制。方法使用Swiss Target Prodiction等数据库中获取烟酸的药效团靶点,再从GEO数据库下载骨性关节炎相关数据集,采用仙桃在线数据分析网站进行差异基因分析,并与烟酸药效团靶点相映射,从String网站中构建PPI互作网络,采用Cytoscape V3.10.1获取Hub gene。结果找到12548个骨性关节炎相关性基因,其中上调6655个,下调5893个;烟酸与骨性关节炎交集基因是8个,富集的生物学功能包括氧化、过氧化、抗氧化和还原功能,富集通路结果包括脂肪细胞中脂肪分解的调节、花生四烯酸代谢、NF-κB信号通路、含血清素的神经突触。结论烟酸治疗骨性关节炎的相关分子功能和药理靶点具有靶点、多通路特点。Objective To discuss the possible mechanism of niacin in the treatment of osteoarthritis by using GEO database combined with network pharmacology.Methods Swiss Target Prodiction and other databases were used to obtain the pharmacophore targets of niacin,and then the osteoarthritis-related data sets were downloaded from the GEO database.The online data analysis website of Xiantao was used for differential gene analysis,and mapped with the pharmacophore targets of niacin.The PPI interaction network was constructed from the String website,and the Hub gene was obtained by Cytoscape V3.10.1.Results A total of 12548 osteoarthritis-related genes were found,of which 6655 were up-regulated and 5893 were down-regulated.There were 8 genes in the intersection of niacin and osteoarthritis.The enriched biological functions included oxidation,peroxidation,antioxidant and reducing functions.The enriched pathway results included the regulation of lipolysis in adipocytes,arachidonic acid metabolism,NF-κB signaling pathway,and serotonin-containing synapses.Conclusion The related molecular functions and pharmacological targets of niacin in the treatment of osteoarthritis have the characteristics of targets and multiple pathways.
