摘  要:目的:探究黄芪多糖对视网膜细胞的保护作用。方法:C57小鼠随机分为对照组、N-甲基-D-天冬氨酸(NMDA)组和黄芪多糖组。对照组和NMDA组小鼠玻璃体腔分别注射磷酸盐缓冲盐水(PBS)和NMDA,并灌胃生理盐水。黄芪多糖组小鼠在注射NMDA后,灌胃黄芪多糖。视网膜铺片和HE染色观察神经节细胞和视网膜厚度变化。视网膜电图(ERG)和视动反应检测视网膜功能和视敏度。结果:黄芪多糖组中,神经节细胞和双极细胞数量较NMDA组增多,且HE显示视网膜厚度增加。ERG和视动反应表明黄芪多糖可以改善视网膜功能和视敏度。结论:黄芪多糖抵抗NMDA诱导模型引起的神经节细胞和双极细胞损伤,改善视网膜结构和功能。Objective:To investigate the protective effect of Astragalus polysaccharide on retinal cells.Methods:Healthy male C57/BL6 mice were randomly divided into the control group,the N-methyl-D-aspartic acid(NMDA)group and the Astragalus polysaccharide group.In the control and NMDA groups,PBS and NMDA were injected intravitreally,respectively.Mice in these groups were then gavaged with saline for 7 days.In contrast,the Astragalus polysaccharide group received Astragalus polysaccharide by gavage following NMDA injection.Retinal flat-mount and hematoxylin-eosin staining(HE)were used to observe the changes in the retina.The function of the mouse retina was measured using electroretinogram(ERG)and optomotor response(OMR).Results:In the Astragalus polysaccharide group,retinal flat-mount and HE staining revealed a significant increase in retinal ganglion cells and thickness compared with the NMDA group.ERG results showed that the Astragalus polysaccharide could restore the retinal function injured by NMDA.Compared with that in the NMDA group,the visual acuity of mice in the Astragalus polysaccharide group was significantly improved.Conclusion:Astragalus polysaccharide protects ganglion and bipolar cells and improves retinal structure and function against NMDA-induced retinal damage.
