摘  要:为探索α-扩张蛋白(α-expansin,EXPA)基因参与铝毒害下对植物生长发育的调控作用,本研究分析了铝处理对地毯草(Axonopus compressus)生长的影响,克隆了地毯草AcEXPA1基因,并进行了生物信息学和基因表达分析。结果表明:与不加铝相比,铝处理抑制了地毯草根系生长,降低了总根长、根表面积和根体积。地毯草AcEXPA1基因全长为774 bp,编码257个氨基酸残基,蛋白分子量为27.7 kDa。AcEXPA1蛋白具有信号肽,被预测定位于胞外基质。实时定量PCR结果表明,AcEXPA1基因在地毯草根中的表达量高于叶中的表达量,且在距离根尖1~2 cm根段中的表达量最高。铝处理增强了AcEXPA1基因在地毯草根中的表达,但镉和镧处理对基因表达无显著影响。不同铝浓度和时间处理结果进一步表明铝增强了AcEXPA1基因的表达。本研究结果为解析地毯草适应铝毒害的机理提供了候选基因资源。To explore the roles ofα-expansin(EXPA)gene in regulation of plant growth and development under aluminum(Al)toxicity,the effects of Al treatments on the growth of carpetgrass(Axonopus compres⁃sus)were analyzed.And AcEXPA1 was cloned for bioinformatic and gene expression analyses.The results showed that Al toxicity inhibited carpetgrass root growth,as reflected by reduced total root length,root surface area and root volume,compared to the control without Al.The full length of AcEXPA1 is 774 bp,encoding 257 amino acid residues.Molecular weight of AcEXPA1 is 27.7 kDa.The AcEXPA1 protein includes a signal peptide and was predicted to be localized to the extracellular matrix.Real-time quantitative PCR analysis showed that AcEXPA1 exhibited higher expression in roots than in leaves,and AcEXPA1 mainly expressed in 1~2 cm root segment away from the root tip.The expression of AcEXPA1 was significantly enhanced in root of carpetgrass by Al but not cadmium and lanthanum treatments.Moreover,the results from different Al concentration and duration treatment confirmed that Al enhanced the expression of AcEXPA1 in carpetgrass roots.This study provides a candidate gene resource for dissecting the Al tolerance mechanisms in carpetgrass.
