南昌市郊黄瓜棒孢叶斑病病原菌分离鉴定

摘  要:以南昌市郊具有典型棒孢叶斑病症状的黄瓜病叶为试材,采用组织分离法、致病性测定、培养性状和形态特征观测及分子生物学鉴定等方法,研究了南昌市郊黄瓜棒孢叶斑病病原菌种类,以期为黄瓜棒孢叶斑病的田间快速诊断和科学防治提供参考依据。结果表明:从黄瓜病叶中共分离获得32个培养性状和形态特征一致的菌株,其对黄瓜叶片均具有致病性。菌株在PDA培养基上的菌落正面灰白色至灰褐色,背面黄褐色,菌丝体致密,绒毛状。菌落平均生长速率10.0 mm·d^(-1)。分生孢子梗粗壮,直立或稍弯曲,单生,浅褐色,有隔膜;分生孢子圆柱形、倒棍棒形、纺锤形或“Y”字形,有多个假隔膜,大小为(22.8~137.8)μm×(4.1~12.9)μm,常单生,偶串生。菌株培养性状和形态特征与已报道的多主棒孢霉(Corynespora cassiicola)相符。3个代表性菌株具有完全相同的rDNA-ITS、ACT和EF-1α基因序列,且与GenBank中C.cassiicola对应序列的同源性均为100%。在基于rDNA-ITS序列构建的系统发育树中,代表菌株与多主棒孢霉(C.cassiicola)菌株聚为同一支,且支持率为100%。根据形态学特征和基因序列分析,将南昌市郊黄瓜棒孢叶斑病病原菌鉴定为多主棒孢霉(C.cassiicola)。The cucumber leaves with typical symptoms of Corynespora leaf spot disease from the suburbs of Nanchang were used as experimental materials.The methods such as tissue isolation,pathogenicity assays,observation of cultural characteristics and morphological traits,and molecular biological identification of the pathogen were used,the pathogen species of cucumber Corynespora leaf spot disease in the suburbs of Nanchang were studied,in order to provide a reference for rapid field diagnosis and scientific control of cucumber Corynespora leaf spot disease.The results showed that a total of 32 isolates with consistent cultural features and morphological characteristics were obtained from diseased cucumber leaves,which were all pathogenic to cucumber leaves.On PDA culture media,colonies of the tested strains were grayish white to grayish brown on the front side,and yellowish brown on the back side.The mycelia were dense and fluffy,with an average growth rate of 10 mm·d^(-1).The conidiophore were solitary,stout,erect or slightly bent,light brown with septa.The conidia were cylindrical,inverted rod-shaped,spindle or‘Y’shape,with multiple pseudo-septa,(22.8-137.8)μm×(4.1-12.9)μm in size,often solitary and occasionally concatenated.The cultural features and morphological characteristics of the strains were consistent with Corynesporacassiicola in the literature.Three representative strains had identical seqences of rDNA-ITS,ACT and EF-1α genes,were 100%identity to the corresponding sequences of C.cassiicola in GenBank.In the phylogenetic tree based on the rDNA-ITS gene sequences,the representative strains was clustered into the same clade as the strains of C.cassiicola with 100%support.According to the morphological characteristics and analysis of the gene sequences,the pathogen of cucumber Corynespora leaf spot in suburbs of Nanchang was identified as C.cassiicola.
