摘 要:为了揭示冷刺激下lncRNA5791的应答模式,本研究利用实时荧光定量PCR技术(qRT-PCR)、核质分离技术、RNA沉降(Pull down)技术全面分析lncRNA5791生物功能。结果表明,lncRNA5791在冷刺激处理的民猪背部脂肪和腹股沟脂肪中相对表达量较高,在臀部脂肪中相对表达量较低。冷刺激处理后猪不同部位脂肪中lncRNA5791的相对表达量均极显著高于常温对照(P<0.01)。lncRNA5791定位于猪的9号染色体,其在细胞核和细胞质中均有分布。在脂肪细胞增殖期,lncRNA5791的表达持续受到抑制;在脂肪细胞分化期,lncRNA5791的相对表达量呈现先升高后下降的趋势。质谱分析结果表明,lncRNA5791可能与膜联蛋白A2(ANXA2)、泛素A52残留核糖体蛋白融合产物1(UBA52)和组蛋白H4(H4)互作。本研究结果为揭示lncRNA5791在冷刺激应答中的具体调控机制奠定了重要基础。In order to reveal the response pattern of lncRNA5791 under cold stimulation,this study comprehensively analyzed the biological functions of lncRNA5791 by using real-time fluorescence quantitative PCR technology(qRT-PCR),nuclear-cytoplasmic separation technology and RNA pull-down technology.The results showed that the relative expression level of lncRNA5791 was relatively high in the back fat and inguinal fat of Min pigs treated with cold stimulation,and relatively low in the gluteal fat.The relative expression levels of lncRNA5791 in the fats of different parts of pigs treated with cold stimulation were all extremely significantly higher than those of the normal temperature control group(P<0.01).lncRNA5791 was located on chromosome 9 of pigs and was distributed in both the nucleus and the cytoplasm.During the proliferation period of adipocytes,the expression of lncRNA5791 was continuously inhibited.During the differentiation period of adipocytes,the relative expression level of lncRNA5791 showed a trend of increasing first and then decreasing.The results of mass spectrometry analysis indicated that lncRNA5791 might interact with annexin A2(ANXA2),ubiquitin A52 residue ribosomal protein fusion product 1(UBA52) and histone H4(H4).The results of this study lay an important foundation for revealing the specific regulatory mechanism of lncRNA5791 in the response to cold stimulation.
