摘  要:利用乙醚去除烟梗中的脂溶性物质,无水乙醇去除可溶性糖,再利用复合酶酶解去除纤维素、果胶、蛋白质等大分子物质;经过二甲基亚砜(DMSO)溶剂溶解提取,利用无水乙醇沉淀淀粉;经沸水复溶后,利用碘-碘化钾溶液显色,经过紫外可见光谱分析,提取的烟梗淀粉纯度达到83.9%。通过优化凝胶渗透色谱(GPC)条件,对烟梗淀粉的分子量进行分析,优化的GPC的分析条件为:样品浓度5.0 mg·m L^(-1)、LS能量25%、柱温40℃、流速1.0 mL·min^(-1),流动相50 mmol·L^(-1)的NaNO3溶液;利用标准品聚氧化乙烯(PEO)对系统校正,测得的烟梗淀粉的分子量分布范围为106~10~8 Da,数均分子量为(Mn)为6.13×10^(6) Da。研究建立了一种有效测定烟梗中淀粉分子量的方法,为分析烟梗的品质提供了重要参考。The lipid soluble substances in the starch of tobacco stems were removed by ether,the soluble sugars were removed by ethanol absolute,and the macromolecules such as cellulose,pectin and protein were removed by enzymolysis;then the starch in tobacco stems was extracted through dimethyl sulfoxide(DMSO)dissolution and ethanol absolute precipitation;after resolubilization in boiling water and colour rendering by iodine-potassium iodide solution,the purity of the extracted tobacco stem starch reached 83.9%as analyzed by UV-visible spectrum.The molecular weight of starch from tobacco stems was analyzed by optimizing the gel permeation chromatography(GPC)conditions.The optimized analytical conditions for GPC were 5.0 mg·m L^(-1) sample concentration,LS energy 25%,column temperature 40℃,flow rate 1.0mL·min^(-1),and mobile phase 50 mmol·L^(-1) NaNO,solution;using polyethylene oxide(PEO)to calibrate the system,the measured molecular weight of tobacco stem starch was 10°~10"Da,and the molecular weight(M,)was 6.13×10^(6) Da.This method established an effective method to measure the molecular weight of starch in tobacco stems and provided an important reference for determining the quality of tobacco stems.
