摘 要:目的:构建小鼠肝组织中脂肪酸(FFA)含量的靶向检测方法,并进行方法学考察,为大黄素的临床应用提供科学数据。方法:将32只小鼠随机分为空白组(C组)、大黄素低剂量组(C_L组)、大黄素中剂量组(C_M组)、大黄素高剂量组(C_H组),每组8只。大黄素给药剂量分别为40 mg/kg,80 mg/kg,120 mg/kg,1次/d,连续给药4周。采用超高效液相色谱-质谱(UHPLC-MS)技术检测小鼠肝组织中FFA含量的变化,利用SIMCA 14.1软件对原始数据进行主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA),比较FFA含量的组间差异,根据变量权重值(VIP)>1.0筛选差异代谢物,通过热图进行可视化分析,明确大黄素对小鼠FFA代谢的影响。结果:C_L组小鼠肝组织中FFA含量与C组比较,差异无统计学意义(P>0.05);而C_M组与C_H组小鼠肝组织中亚油酸[FFA(18∶2)]、花生四烯酸[FFA(20∶4)]含量明显高于C组,差异有统计学意义(P<0.01),即C_M组与C_H组小鼠肝组织中Omega-6多不饱和脂肪酸(n-6 PUFAs)产生堆积。与C组比较,C_L组、C_M组与C_H组小鼠肝组织中FFA(20∶4)/二十二碳六烯酸[FFA(22∶6)]、棕榈酸[FFA(16∶0)]/FFA(22∶6)升高,差异有统计学意义(P<0.01)。结论:FFA(18∶2)、FFA(18∶0)、FFA(18∶1)、FFA(16∶0)、FFA(20∶4)、FFA(20∶4)/FFA(22∶6)、FFA(16∶0)/FFA(22∶6)可能是大黄素调节肝组织FFA代谢的潜在生物标志物。Objective:To establish a targeted detection method for fatty acid(FFA)content in mouse liver tissues,investigate the methodology,and provide scientific data for the clinical application of emodin.Methods:32 mice were randomly divided into blank group(C),low-dose emodin group(C_L),medium-dose emodin group(C_M),high-dose emodin group(C_H),with 8 mice in each group.Emodin was administered at 40 mg/kg,80 mg/kg and 120 mg/kg once a day for 4 weeks.Ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS)was used to detect the changes of FFA content in mouse liver tissues.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were performed on the original data using SIMCA 14.1 software to compare the difference of FFA content between groups.Differential metabolites were screened according to variable importance projection(VIP)>1.0.The effects of emodin on FFA metabolism in mice were visualized by heat map analysis.Results:There was no statistical significance in FFA content in mouse liver tissues between C_L group and C group(P>0.05).The contents of Linoleic acid[FFA(18:2)]and arachidonic acid[FFA(20:4)]in mouse liver tissues in C_M group and C_H group were significantly higher than those in C group,and the difference was statistically significant(P<0.01),which indicated that Omega-6 polyunsaturated fatty acids(n-6 PUFAs)were accumulated in mouse liver tissues in C_M and C_H groups.Compared with those of C group,FFA(20:4)/docosahexaenoic acid[FFA(22:6)]and palmitic acid[FFA(16:0)]/FFA(22:6)in mouse liver tissue in C_L group,C_M group and C_H group were increased,and the differences were statistically significant(P<0.01).Con⁃clusion:FFA(18:2),FFA(18:0),FFA(18:1),FFA(16:0),FFA(20:4),FFA(20:4)/FFA(22:6),and FFA(16:0)/FFA(22:6)may be potential biomarkers of emodin regulation of FFA metabolism in liver tissues.
