摘 要:目的:采用秀丽隐杆线虫模型研究淫羊藿苷的抗衰老活性及作用机制。方法:通过测定淫羊藿苷对铜绿假单胞菌PA14侵染线虫的生存率、活力和体内脂肪含量的影响以及对野生型N2线虫氧化应激、热应激和脂褐素的影响探讨淫羊藿苷的抗衰老活性,通过测定突变体线虫TJ356衰变加速因子-16(decayacceleratingfactor-16,DAF-16)的蛋白核转位和突变体线虫CF1553体内超氧化物歧化酶-3(superoxide dismutase-3,SOD-3)的表达探究淫羊藿苷抗衰老的分子机制。结果:淫羊藿苷显著提高了在铜绿假单胞菌PA14侵染下线虫的生存率、活力和体内脂肪含量,增强了在热应激损伤和氧化应激损伤下线虫的耐受性,降低了脂褐素沉积。淫羊藿苷能够促进突变体线虫TJ356的DAF-16核转位,显著提升突变体线虫CF1553体内SOD-3蛋白的表达水平。结论:淫羊藿苷具有显著的抗衰老活性,可能通过促进DAF-16核转位、激发SOD-3的表达实现对线虫的保护作用。Objective:To study the anti-aging activity and mechanism of icariin by using Caenorhabditis elegans models.Methods:The anti-aging activity of icariin was investigated by determining its effects on the survival rate,vitality,and body fat content of Caenorhabditis elegans infected with Pseudomonas aeruginosa PA14,as well as its impact on the oxidative stress,heat stress,and lipofuscin levels in wild-type N2 Caenorhabditis elegans.Additionally,to explore the molecular mechanism underlying the anti-aging properties of icariin,the nuclear translocation of decay accelerating factor-16(DAF-16)in mutant nematode TJ356 and the expression of superoxide dismutase-3(SOD-3)in mutant nematode CF1553 were measured.Results:Icariin significantly improved the survival rate,vitality,and body fat content of Caenorhabditis elegans in P.aeruginosa PA14 infection,enhanced the tolerance of Caenorhabditis elegans under heat stress and oxidative stress injury,decreased the deposition of lipofuscin,and promoted the DAF-16 nuclear translocation of mutant Caenorhabditis elegans TJ356.The expression level of SOD-3 protein in mutant nematode CF1553 was significantly increased.Conclusion:Icariin has significant anti-aging activity which is probably achieved by promoting the nuclear translocation of DAF-16 and stimulating the expression of SOD-3.
