摘  要:目的分析环氧化酶2(Cyclooxygenase-2,COX2)在肥胖、阻塞性睡眠呼吸暂停综合征(obstructive sleep apnea-hypopnea syndrome,OSAHS)幼鼠肝、肾、心、脑组织中的表达与定位情况,为儿童肥胖及OSAHS的早期识别及靶向治疗提供理论依据。方法24只3~4周龄雄性Wistar幼鼠随机分为Control组(A组)、Obesity组(B组)、OSAHS组(C组)、Obesity+OSAHS组(D组)。B组及D组用高脂饲料进行喂养,A组及C组用普通饲料喂养;C组及D组通过定时电磁阀将纯氮气和压缩空气分配到各个腔室构建OSAHS模型。运用Western Blot方法对幼鼠肝、肾、心、脑组织中COX2的表达水平进行检测;运用免疫组织化学法对幼鼠肝、肾、心、脑组织中COX2的表达及定位进行检测。结果B组和D组幼鼠体重、总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)含量与A组相比均显著增加(P<0.05),C组和D组幼鼠血氧饱和度与A组相比均显著降低(P<0.05)。B组、C组、D组幼鼠肝、肾、心、脑组织中COX2表达水平较A组均显著升高(P<0.05),D组幼鼠肝、肾、心、脑组织中COX2表达水平较B组和C组均显著升高(P<0.05)。结论COX2在肥胖OSAHS幼鼠肝、肾、心、脑组织中均有表达,其中肝组织敏感性较高,脑组织敏感性较低。Objective To analyze the expression and localization of COX2 in different tissues of young obese OSAHS rats,and to provide a theoretical basis for early identification and targeted treatment of childhood obesity and OSAHS.Methods Twenty-four 3-4 weeks young rats were randomly divided into the Control group(A group),the Obesity group(B group),the OSAHS group(C group),and Obesity+OSAHS group(D group).The A group and the C group were fed with normal rat chow,the B group and the D group were fed with high-fat rat chow,and the C group and the D group distributed pure nitrogen and compressed air to the chambers via timed solenoid valves to establish animal models.We used Western Blot and immunohistochemistry to test the expression levels and the distribution of COX2 in the liver,kidney,heart,and brain tissues of young rats.Results The body weight,total cholesterol,and triglyceride content of the young rats in the B group and the D group were significantly increased compared with the A group(P<0.05),and the oxygen saturation of the young rats in the C group and the D group was significantly decreased compared with the A group(P<0.05).The expression levels of COX2 in the liver,kidney,heart,and brain tissues of young rats in the B,C and D groups were significantly higher than those in the A group(P<0.05),and the expression levels of COX2 in liver,kidney,heart and brain tissues of young rats in the D group were significantly higher than those in the B group and the C group(P<0.05).Conclusion COX2 is expressed in the liver,kidney,heart,and brain tissues of young obese OSAHS rats,with higher sensitivity in the liver tissue,and lower sensitivity in the brain tissue.
