摘 要:为筛选miR-182-5p影响籽鹅精子迁移率性状的关键靶基因,首先利用在线数据库预测miR-182-5p的靶基因,然后进行GO功能注释、KEGG通路注释和蛋白互作分析,并利用Cytoscape软件绘制靶基因可能参与的调控网络。最后,利用RNA测序技术分析这些靶基因在1岁龄籽鹅睾丸中的表达,根据microRNA与靶基因负调控的作用机制筛选关键靶基因。结果表明:共获得97个候选靶基因;GO富集分析显示,这些靶基因涉及细胞骨架、GTPase调控、大分子结合和代谢过程;KEGG富集分析显示,PI3K-Akt、钙、Rap1等信号通路被显著富集(P<0.05);蛋白互作网络筛选出32个基因存在互作;表达谱分析共筛选出6个基因,包括RECK、BROX、CUL5、LSM14A、TNFILR1和SIPA1L2。因此,miR-182-5p通过调节FGF9、CUL4、TRAILR、SIPA1L2和RECK的表达,对睾丸发育和精子发生发挥积极影响。In order to identify the key target genes of miR-182-5p affecting sperm mobility traits in Zi geese,the online databases were used to predict the target genes of miR-182-5p.Subsequently,GO functional annotation,KEGG pathway annotation and protein interaction analysis were performed,and Cytoscape software was used to map the regulatory network of target genes.Finally,the expression of these target genes in the tests of one-year-old geese were analyzed by RNA sequencing technology,and the key target genes were identified based on the mechanism of negative correlation between microRNA and target genes.The results showed that:a total of 97 candidate target genes were identified;GO enrichment analysis showed that these target genes were involved in cytoskeleton,GTPase regulation,macromolecular binding and metabolic processes;KEGG enrichment analysis showed that PI3K-Akt,calcium,Rap1 and other signaling pathways were significantly enriched(P<0.05);32 genes were identified by protein interaction network to interact with each other;6 genes were identified by expression analysis,including RECK,BROX,CUL5,LSM14A,TNFILR1 and SIPA1L2.In conclusion,miR-182-5p exerted a positive effect on testicular development and testicular development by regulating the expression of FGF9,CUL4,TRAILR,SIPA1L2 and RECK.
